摘要
目的研究一株植物内生真菌Stagonosporopsis sp.YZHH-J-1的次级代谢产物及其抗菌活性。方法采用正、反相硅胶柱色谱以及高效液相等多种现代分离方法进行对该菌株粗提物分离、纯化;利用质谱(MS)和核磁共振(NMR)等波谱学技术对分离得到的化合物进行结构鉴定;评价单体化合物的体外抗菌活性以及生物被膜形成抑制活性。结果从菌株Stagonosporopsis sp.YZHH-J-1大米固体发酵产物的乙酸乙酯粗提物中共分离鉴定了8个化合物,分别为aspergillol A(1)、阿魏酸(2)、4-羟基苯乙基乙酸酯(3)、对羟基苯乙酸甲酯(4)、吲哚-3-甲醛(5)、octadecyl 3-(3,5-di-tert-butyl-4-hydroxyphenyl)propionate(6)、(9Z,11E)-13-oxo-9,1-octadecadienoic acid(7)和亚油酸(8)。化合物1~8均为首次在Stagonosporopsis属真菌中分离得到;其中,化合物4、7和8具有抑制格特隐球菌活性,MIC值为32~64μg/mL;此外,化合物7和8分别对耐甲氧西林金黄色葡萄球菌(MRSA)和青枯雷尔菌(Ralstonia solanacearum)具有抑制作用,MIC值分别为64μg/mL和32μg/mL。化合物7对MRSA以及化合物8对R.solanacearum的生物被膜形成均具有一定抑制活性。结论植物内生真菌Stagonosporopsis sp.YZHH-J-1能够产生具有一定抗菌活性的次级代谢产物,为抗菌药物先导化合物的发现奠定了基础。
Objective To isolate and identify secondary metabolites from the endophytic fungus Stagonosporopsis sp.YZHH-J-1 and their antimicrobial activity.Methods The crude extract of the strain was separated and purified by various modern separation methods,including normal and reversed-phase silica gel column chromatography and high-performance liquid chromatography.The structures of the compounds were characterized using spectroscopic techniques such as mass spectrometry(MS)and nuclear magnetic resonance(NMR).The in vitro antimicrobial and inhibition of biofilm formation activity of the isolated compounds was evaluated.Results Eight compounds were isolated from the crude extract of Stagonosporopsis sp.YZHH-J-1 and identified as aspergillol A(1),(E)-ferulic acid(2),4-hydroxy phenethyl acetate(3),methyl p-hydroxyphenylacetate(4),indole-3-carboxaldehyde(5),octadecyl 3-(3,5-di-tert-butyl-4-hydroxyphenyl)propionate(6),(9Z,11E)-13-oxo-9,1-octadecadienoic acid(7)and linoleic acid(8),respectively.Compounds 1-8 were isolated for the first time from the fungus Stagonosporopsis spp..Compounds 4,7 and 8 showed inhibitory activity against Cryptococcus gatti with MIC values ranging from 32 to 64μg/mL.In addition,compounds 7 and 8 also exhibited inhibitory effects against methicillin-resistant Staphylococcus aureus(MRSA)and Ralstonia solanacearum,with MIC values of 64 and 32μg/mL,respectively.Compound 7 showed some inhibitory activity against MRSA as well as compound 8 against R.solanacearum biofilm formation.Conclusion The endophytic fungus Stagonosporopsis sp.YZHH-J-1 produced secondary metabolites with antimicrobial activity and provided lead compounds for the development of antimicrobial drug.
