摘要
目的进一步开发利用药用植物华北大戟,对该植物内生真菌进行分离鉴定,初步探索真菌种类的多样性及其抗肿瘤增殖活性。方法采用组织切块法对真菌进行分离培养,利用形态学特征和IST序列分析鉴定菌株种属,利用MEGA 7.0.14软件中的邻接法构建系统发育树。以HepG2细胞为抗肿瘤增殖活性测试细胞株,CCK8法检测菌株粗提物抑制肿瘤细胞增殖的作用。结果从中分离得到9株内生真菌,比对分析确定属于5个属,分别为曲霉属、青霉属、毛壳菌属、链格孢属、蜡蚧菌属。抗肿瘤增殖活性筛选结果显示菌株1、4、5号有抑制HepG2肿瘤细胞增殖的能力,IC 50值为90.7、72.5、80.3μg/mL。结论研究初步确定了华北大戟内生真菌的菌属,为真菌次级代谢产物的后续抗肿瘤增殖活性成分研究提供了基础。
Objective To further develop and utilize the medicinal plant of Euphorbia esula,isolate and identify endophytic fungi from the plant,and preliminarily explore the diversity of fungal species and their anti-tumor proliferation activity of endophytic fungi.Methods The endophytic fungi were isolated and purify by using tissue block isolation.The species of endophytes were preliminarily analyzed by morphological characteristics and then by ITS sequence analysis to determine their taxonomic status.The phylogenetic tree was built by using the adjacency method in MEGA 7.0.14 software.HepG2 cells were used as the anti-tumor proliferation activity test cell line,and the CCK8 method was used to determine the inhibitory effect of the crude extract on tumor cells proliferation.Results Nine endophytic fungi were isolated from endophytic fungi and five genera were identified:Aspergillu,Penicillium,Chaetomium,Alternaria,and Lecanicillium.The screening results of anti-tumor proliferation activity showed that strains 1,4 and 5 had the ability to inhibit HepG2 tumor cells proliferation,with IC 50 values of 90.7μg/mL,72.5μg/mL and 80.3μg/mL.Conclusion The study preliminarily determined the types of endophytic fungi in Euphorbia esula,providing a specific basis for research on the subsequent study of anti-tumor proliferation active ingredients in secondary metabolites of fungi.
