三七总皂苷对大鼠心肌缺血再灌注损伤的影响及抗焦亡机制研究 被引量:1

Effects of Panax Notoginseng Saponins on Myocardial Ischemia-reperfusion Injury in Rats and the Mechanism of Anti-pyroptosis

摘要 目的:观察三七总皂苷(PNS)对心肌缺血再灌注(IR)损伤的作用,探讨其机制是否与调控心肌细胞焦亡有关。方法:采用随机数字表法将18只成年雄性SD大鼠分为假手术组、IR组和PNS组,各6只。采用结扎大鼠冠状动脉左前降支(LAD)制备模型,缺血30 min,再灌注2 h;假手术组和IR组大鼠腹腔注射生理盐水(10 mL/kg),PNS组于造模前30 min腹腔注射PNS(50 mg/kg)。采用苏木精-伊红(HE)染色观察心肌损伤程度;免疫组化法检测半胱氨酸天冬氨酸特异性蛋白酶-1(Caspase-1)表达;蛋白免疫印记法(Western Blot)检测心肌组织Caspase-1、消化道皮肤素D(GSDMD)蛋白含量的表达。结果:与假手术组比较,IR组心肌损伤程度加重,Caspase-1、GSDMD蛋白表达增加(P<0.05);与IR组比较,PNS组心肌损伤程度减轻,Caspase-1、GSDMD蛋白表达降低(P<0.05)。结论:三七总皂苷可减轻大鼠心肌IR损伤,其作用机制可能与抑制心肌细胞焦亡有关。 Objective:To observe the effect of Panax notoginseng saponins(PNS)on myocardial ischemia-reperfusion(IR)injury and explore its mechanism related to the regulation of cardiomyocyte pyroptosis.Methods:Eighteen adult male SD rats were divided into sham group,IR group and PNS group by random number table method,with 6 rats in each group.The model was prepared by ligating the left anterior descending coronary artery(LAD)of rats,with ischemia for 30 minutes and reperfusion for 2 hours.The rats in the sham group and the IR group were intraperitoneally injected with normal saline(10 mL/kg),while those in the PNS group were intraperitoneally injected with PNS(50 mg/kg)30 minutes before modeling.The degree of myocardial injury was observed by hematoxylin-eosin(HE)staining.The expression of cysteine aspartate specific protease-1(Caspase-1)was detected by immunohistochemistry.The expressions of Caspase-1 and gasdermin D(GSDMD)protein contents in myocardial tissue were detected by Western Blot.Results:Compared with the sham group,the degree of myocardial injury in the IR group was aggravated,and the protein expressions of Caspase-1 and GSDMD increased(P<0.05).Compared with the IR group,the degree of myocardial injury in the PNS group was alleviated,and the protein expressions of Caspase-1 and GSDMD decreased(P<0.05).Conclusion:Panax notoginseng saponins could alleviate myocardial IR injury in rats,and its mechanism might relate to the inhibition of cardiomyocyte pyroptosis.
