小鼠自身免疫性心肌炎心肌ICAM-1的表达变化及三七总皂甙的干预作用 被引量:3

摘要 目的:探讨自身免疫性心肌炎小鼠心肌组织细胞间黏附分子-1(ICAM-1)的表达变化以及三七总皂甙对其的影响。方法:BALB/C雄性小鼠72只,随机均分为对照组(C组)、自身免疫性心肌炎组(A组)和三七总皂甙组(P组)。A组和P组复制自身免疫性心肌炎模型。P组腹腔注射三七总皂甙进行干预。于第14、第21和第30天随机处死各组小鼠8只,留取心脏标本。电镜观察心肌组织超微结构改变,免疫组化法检测心肌组织ICAM-1蛋白表达。结果:A组心肌组织的超微结构损伤严重,P组较A组损伤程度明显减轻;C组心肌组织ICAM-1蛋白表达量极少,A组ICAM-1蛋白表达明显增加,呈动态变化,P组心肌细胞ICAM-1蛋白表达显著低于A组(P<0.05或P<0.01)。结论:自身免疫性心肌炎可诱导心肌组织ICAM-1蛋白的表达,三七总皂甙可能通过下调心肌组织ICAM-1蛋白的表达而减轻心肌损伤。 Objective:To observe the influence of panax notoginseng saponins(PNS) on expression of ICAM-1 in myocardial tissue of mice during autoimmune myocarditis(AMC). Methods: A total of 72 inbred five-week-old male BALB/C mice were randomly divided into three groups: control group(C group), autoimmune myocarditis group(A group) and panax notoginseng saponins group(P group). Group A and group P were immunized with porcine heart myosin (100 μg) in an equal content of CFA in a total volume of 0.2 ml, while C group was injected with PBS in an equal content of CFA. P group were subcutaneously injected with PNS(40 mg/kg) in inguinal groove for 5 days, while C and A group were treated with physiological saline. Eight mice of each group were sacrificed in 14 th, 21 th and 30th day. The ultrastructure changes were observed under electron microscope. The expression of ICAM-1 was measured in myocardial tissue by immunohistochemistry(IHC). Results: Electron microscope showed that the pathological injury of myocardium in P group was obvious attenuated as compared to A group. The IHC demonstrated that ICAM-1 protein expression in P group was significantly lower than that in A group.Conclusion: The expression of ICAM-1 increases during AMC, while Panax notoginseng saponins may have a protective effect in AMC by down-regulating the expression of ICAM-1.
