摘要
利用H_(2)O_(2)处理Caco-2细胞构建氧化损伤模型,检验3条芡实抗氧化肽(WGP为F_(1)、DEPG为F_(2)、PAIRY为F_(3),纯度>98%)对氧化应激细胞保护作用。结果表明,400μmol/L的H_(2)O_(2)处理Caco-2细胞12 h,细胞存活率为(54.72±1.48)%,为构建氧化损伤模型细胞最佳条件;Vc为对照组进行体外抗氧化能力测验,F_(2)对DPPH·、ABTS^(+)·、·OH自由基清除能力IC_(50)值分别为(121.16±3.67)、(61.24±0.62)、(96.47±5.79)μg/m L;细胞毒性试验表明,F_(2)对Caco-2细胞无毒性抑制性,无促进增殖作用;F_(2)浓度为240μg/m L时,Caco-2细胞存活率达到了(94.74±3.69)%,比阳性对照组提高了0.73倍。细胞内MDA含量降低了(57.25±3.27)%,SOD、CAT、GSH-Px酶活分别提高了(345.00±12.07)%、(381.25±13.16)%、(228.79±18.42)%,细胞凋亡率降低到(9.54±0.97)%。研究表明F_(2)可以显著降低氧化损伤细胞的凋亡率,降低细胞的氧化应激损伤、保护细胞,为F_(2)在医药和食品领域高值化开发利用提供了有力的理论依据和试验支撑。
An oxidative stressed induced Caco-2 cells model was established by H_(2)O_(2)treatment,and the cytoprotective effects of three antioxidant peptides from Semen euryales(WGP designated as F_(1),DEPG as F_(2),and PAIRY as F_(3);purity>98%)on the damaged Caco-2 cells were studied.It was found that the oxidative damage Caco-2 cell model was treated with 400μmol/L H_(2)O_(2)for 12 hours,with cell survival rate of(54.72±1.48)%.Vitamin C(Vc)was used as a positive control for in vitro antioxidant capacity testing,and F_(2)exhibited IC_(50)values for DPPH·,ABTS^(+)·,and·OH radical scavenging ability was(121.16±3.67),(61.24±0.62),and(96.47±5.79)μg/mL,respectively.The cytotoxicity test showed that F_(2)had neither toxic inhibitory effect nor proliferative effects on Caco-2 cell and did not promote cell proliferation.When F_(2)concentration was 240μg/mL,the survival rate of Caco-2 cells reached(94.74±3.69)%,which was 73%higher than the positive control group.The intracellular MDA level was decreased by(57.25±3.27)%,while the activities of SOD,CAT,and GSH-Px enzymes were increased by(345.00±12.07)%,(381.25±13.16)%,and(228.79±18.42)%,respectively.The apoptosis rate of the cells was decreased to(9.54±0.97)%.These results demonstrated that F_(2)significantly reduced apoptosis rate of oxidative stress damaged in Caco-2 cells.The findings provide strong theoretical basis and experimental support for the potential high-value development and utilization of F_(2)in the pharmaceutical and food industries.
