摘要
利用H2O2处理HEK293细胞构建细胞氧化损伤模型,研究4条女贞子抗氧化肽(LMVGLT、VPYPHLM、NRDDMRER、ANQLDQFSR)作用于模型细胞,检验其对氧化应激细胞的保护作用。结果表明,400μmol/L的H2O2处理HEK293细胞12h,细胞存活率为(61.34±0.57)%,此条件下构建了最佳的细胞氧化损伤模型;与维生素C(VC)作为对照组进行体外抗氧化能力测验,VC对DPPH自由基、ABTS+自由基、超氧阴离子自由基、羟基自由基的清除率显著高于女贞子抗氧化肽,而抗氧化肽对亚铁离子及铜离子螯合率显著高于VC;细胞毒性试验表明,4条抗氧化肽对HEK293细胞无毒性抑制性,也无促进增殖作用;其中肽2(VPYPHLM)在浓度为240μmol/mL时,HEK293细胞存活率达到了98.98%。细胞中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的酶活和还原型谷胱甘肽(GSH)的含量及GSH/GSSG比值比阳性对照组明显上升,丙二醛(MDA)含量明显下降。研究结果显示女贞子抗氧化肽对氧化损伤细胞具有显著的保护作用,为女贞子抗氧化肽在医药和食品领域高值化开发利用提供了有力的理论依据和试验支撑。
Using H2O2 to treat HEK293 cells for the construction of oxidative damage model cells to study the effects of 4 Ligustrum lucidum antioxidant peptides(LMVGLT,VPYPHLM,NRDDMRER,ANQLDQFSR)on model cells,its protective effect on oxidative stress cells was tested.The results showed that the survival rate of HEK293 cells treated with 400μmol/L H2O2 for 12 h was(61.34±0.57)%,indicating the construction of the optimal oxidative damage model cells under this condition;In vitro antioxidant capacity tests were conducted with vitamin C(VC)as the control group.VC showed significantly higher scavenging rates of DPPH free radicals,ABTS+free radicals,superoxide anion free radicals,and hydroxyl free radicals compared to Ligustrum lucidum antioxidant peptides,while antioxidant peptides had significantly higher chelating rates of ferrous and copper ions than VC;Cell toxicity experiments showed that the 4 antioxidant peptides had no toxic inhibitory effect on HEK293 cells,nor did they promote proliferation;Among them,peptide 2(VPYPHLM)achieved a survival rate of 98.98%of HEK293 cells at the concentration of 240μmol/L.In cells,the content of catalase(CAT),superoxide dismutase(SOD)and reduced glutathione(GSH)and the GSH/GSSG ratio increased obviously compared to the positive control group,while malondialdehyde(MDA)decreased obviously.The research results showed that the antioxidant peptides of Ligustrum lucidum have a significant protective effect on cells with oxidative damage,providing strong theoretical basis and experimental support for the high-value development and utilization of Ligustrum lucidum antioxidant peptides in the fields of medicine and food.
