摘要
[目的]研究梵净山石斛林下仿野生抚育的生长特性及测定其多糖和甘露糖含量。[方法]采用林下吊盆、附树和附石栽培模拟梵净山石斛野生生境生长,利用苯酚-硫酸法测定梵净山石斛多糖含量,柱前衍生化-超高效液相色谱法(UPLC)测定甘露糖含量。[结果]林下吊盆抚育梵净山石斛株高、鲜重、干重显著高于附石、附树抚育(P<0.05);吊盆抚育梵净山石斛多糖和甘露糖含量分别为20.41%、14.23%,与附石、附树梵净山石斛比较,吊盆抚育梵净山石斛多糖和甘露糖含量均具有显著差异(P<0.05)。[结论]林下吊盆抚育是梵净山石斛仿野生抚育及优质高产栽培的有效方式。
[Objective]To study the growth characteristics of Dendrobium fanjingshanense from imitation wild tending in the forest and determine the content of polysaccharide and mannose.[Method]Using hanging pots,attached trees and attached stones under the forest to simulate the wild habitat growth of Dendrobium fanjingshanense,the content of polysaccharides in Dendrobium fanjingshanense was determined by phenol-sulfuric acid method,and the mannose content was determined by pre column derivatization-ultra-high performance liquid chromatography(UPLC).[Result]The height,fresh weight and dry weight of Dendrobium fanjingshanense grown in hanging pots under the forest were significantly higher than those grown in attached stones and trees(P<0.05).The polysaccharide and mannose contents of Dendrobium fanjingshanense grown in hanging pots were 20.41%and 14.23%,respectively.Compared with Dendrobium fanjingshanense grown in attached stones and trees,the polysaccharide and mannose contents of Dendrobium fanjingshanense grown in hanging pots were significantly different(P<0.05).[Conclusion]Hanging pots under the forest for nurturing is an effective way to imitation wild tending and cultivate high-quality and high-yielding of Dendrobium fanjingshanense.
