顺铂-乙基纤维素微囊犬肝动脉栓塞的实验研究

EXPERIMENTAL STUDY OF EMBOLIZATION ON CANINE HEPATIC ARTERY WITH CISPLATIN-ETHYLCELLULOSE MICROCAPSULE

摘要 本文报告用顺铂-乙基纤维素微囊进行犬肝动脉栓塞的实验,观察肝脏病理改变、肝功改变、X线血管造影改变及药代动力学改变的结果,为临床应用提供参考依据。 In this study, CDDP-mc containing 32.5% (w/w) CDDP had been infused into canine hepatic artery (n=6) and compared with the control group (n=6) infused with CDDP alone. Angiograph showed disapearance of the peripheral small branches of the hepatic artery in the tested group and no evidence of collaterals and recanalization 20 days after infusion. Histologic findings of the liver showed local sludging of blood, focal degeneration and necrosis. GPT elevated transiently in the first 3 days. Pharmacokinetic analysis revealed that T 1/2β was prolonged in the tested group nearly as 3.8 times as that of the control group. All these results suggest that CDDP-mc could have not only the function of inducing peripheral embolization, but also prolonging drug release at the same time, and besides, the exposure of the drug to the systemic circulaton also decreased. Thus CDDP-mc could be an available chemoembolus for hepatic embolization in case of hepatic cancer clinically.
