高原鼢鼠肌肉脂溶性物质的抗缺氧效果与化学成分 被引量:10

摘要 为了确定高原鼢鼠 (Myospalaxbaileyi)体内抗缺氧成分 ,通过超临界萃取 (SCEF)方法从高原鼢鼠肌肉提取脂溶性部分 ,我们对其成分通过气相色谱 -质谱 (GC MS)联用分析。为了证实高原鼢鼠肌肉脂溶性成分的抗缺氧效果 ,探讨其抗缺氧机制 ,我们以小鼠为实验动物 ,分为 5个组 ,即高剂量实验组 (HEG)、中剂量实验组 (MEG)、低剂量实验组 (LEG)、阳性药物对照组 (PEG)、空白对照组 (CEG) ,分别以浓度为 2 0 %萃取物、 10 %萃取物、 5 %萃取物、 10 %的红景天和蒸馏水连续饲喂 10天 ,然后进行小鼠常压抗缺氧实验 ,测定小鼠血清超氧化物歧化酶 (SOD)、谷光甘肽过氧化物酶 (GSH PX)、乳酸脱氢酶 (LDH)活性及血清丙二醛(MAD)含量。结果发现 ,高原鼢鼠肌肉脂溶性物质提取率为 11 5 %,共确定了 17种成分 ,主要成分是n -十六烷酸 (32 2 93%)、n -十七烷酸 (1 10 9%)、n -十八烷酸 (6 0 3%)、二 -羟基 -环十五酮 (1 198%)、顺十六烯酸 (6 13%)、反十八烯酸 (2 7 3%)和亚油酸 (18 4 %)。小鼠抗缺氧实验和生化指标测试结果表明 ,高原鼢鼠肌肉脂溶性成分在 2 0 %、 10 %、 5 %时 ,与空白对照组相比 ,都不同程度地延长小鼠在缺氧条件下的存活时间 ,提高小鼠血清SOD和GSH PX活性 ,降低血清LDH活性 。 The plateau zokor is a burrowing rodent of high altitude areas. The oxygen content in their burrows can be as much as 20% less than that of the surface air. To identify anti hypoxic ingredients in plateau zokor muscle the fat soluble component was extracted by means of supercritical fluid extraction (SCEF), and the ingredients were analyzed by gas chromatography mass spectrometry (GC MS). To test the anti hypoxic effects and investigate the mechanisms involved, one hundred mice were randomly divided into five groups: a high dose experimental group (HEG), a medium dose experimental group (MEG), a low dose experimental group (LEG), a positive control group (PCG) and a negative control group (NCG). The HEG, MEG and LEG groups were fed with 20%, 10% and 5% of the fat soluble component respectively, the positive control group was fed with 10% of Hong jingtian (an anti hypoxic traditional Chinese medicine), and the negative control group were fed with distilled water. Each group was fed its respective treatment for ten days,each animal receiving a constant daily dose of 20 ml/kg body weight. Experiments were conducted at normal pressure. The survival time, and metabolic activity of superoxide dismutase (SOD), glutathione peroxidase (GSH PX), lactate dedydrognase (LDH) and the contents of malondialdehyde (MDA) in mouse serum were determined. The results showed that the extraction ratio of the fat soluble component was 11 5%. Seventeen kinds of ingredients were identified, the main ones being hexadecanoic acid (32 293%), heptadecanoic acid (1 109%), octadecanoic acid (6 03%), cyclopentadecanoic, 2 hydroxy (1 198%), Z hexadecenoic acid (6 13%), E octadecenoic acid (27 3%) and Z,Z octadecenoic acid (18 4%). Compared with the negative control group, the fat soluble component significantly prolonged the survival time of mice under hypoxic conditions ( P <0 05), increased SOD and GSH PX activity and markedly ( P <0 05)decreased MDA serum content when provided at concentrations of 10% and 20%. Five percent of the fat soluble component had no effects on SOD, GSH Px and LDH activity but significantly( P <0 05)decreased MDA serum content. These results indicate that the survival time of mice under hypoxic conditions, enzyme activity and MDA serum content were no different between the 10% fat soluble component group and 10% Hong jingtian group. In conclusion, the fat soluble component had significant anti hypoxic function at a concentration of 10%.
