摘要
为研究慢性冷暴露条件下小鼠结肠紧密连接的变化,试验将小鼠随机分为对照组和冷暴露组,对照组始终置于25℃环境中,冷暴露组置于4℃环境中每天暴露3 h,持续刺激3周,其余时间置于25℃环境中。冷暴露周期结束后将小鼠麻醉处死。利用HE染色观察小鼠结肠的形态学变化,免疫荧光染色观察ZO-1的表达,蛋白免疫印迹技术检测紧密连接相关蛋白Claudin-1、Occludin、ZO-1和炎症因子TNF-α、IL-1β、iNOS的表达。结果显示,与对照组相比,冷暴露组小鼠结肠皱襞消失,紧密连接蛋白的表达量显著降低,炎症因子表达量上调。上述结果表明,慢性冷暴露会导致小鼠结肠紧密连接受损,同时促进炎症反应的发生。
To study the changes of colonic tight junctions in mice under chronic cold exposure,mice were randomly divided into control group and cold exposure group.The control group was kept in normal atmospheric temperature(25℃).Mice in the cold exposure group were exposed to 4℃for 3 h every day and were kept in 25℃environment for the rest of the time and the stimulation last for 3 weeks.The mice were anesthetized and killed after cold exposure.The morphological changes of colon were observed by HE staining.The expression level of ZO-1 was observed by immunofluorescence.The expression of tight junction related proteins Claudin-1,Occludin and ZO-1,and inflammatory factors TNF-α,IL-1βand iNOS were detected by Western blot.The results showed that compared with the control group,colonic folds disappeared in cold-exposed mice,the expression of tight junction protein decreased,and the expression of inflammatory factors increased.The results showed that cold exposure resulted in impaired colonic?s tight junctions in mice and promoted inflammation.
