别旁茶通过上调CAV-1对炎症性肠病细胞模型增殖、凋亡的影响 被引量:2

摘要 目的 探讨别旁茶是否通过调控小窝蛋白-1(caveolin-1,CAV1)的表达影响炎症性肠病(IBD)细胞模型的增殖和凋亡。方法 采用RT-qPCR和蛋白免疫印迹验证CAV-1-siRNA的敲低效率和pcDNA-CAV-1的过表达效率。采用CCK-8测定别旁茶处理对各组HT-29细胞的增殖影响。蛋白免疫印迹检测别旁茶处理对各组凋亡相关蛋白的表达。蛋白免疫印迹检测pcDNA-CAV-1和别旁茶处理对Occludin和Claudin-1的蛋白表达水平的影响。结果 在HT-29细胞中CAV-1-siRNA的敲低效率和pcDNA-CAV-1的过表达效率得到验证。别旁茶可以通过上调CAV-1促进IBD细胞模型中HT-29细胞的增殖水平和抗凋亡蛋白的表达。别旁茶可以通过上调CAV-1来促进Occludin和Claudin-1的蛋白表达水平。结论 别旁茶通过上调CAV-1来促进Occludin和Claudin-1的蛋白表达水平,从而促进HT-29细胞的增殖水平并抑制其凋亡水平,在IBD细胞模型中发挥保护作用。 Objective To explore whether Bie Pang Cha(BPC,Elongate Jasmine twig and leaf) affects the proliferation and apoptosis of inflammatory bowel disease cell model by regulating the expression of Caveolin-1(CAV-1). Methods RT-qPCR and Western Blot were used to verify the knockdown efficiency of CAV-1-siRNA and the overexpression efficiency of pcDNA-CAV-1. CCK-8 was used to determine the effect of Jasmine on the proliferation of HT-29 cells in each group. Western Blot method was used to detect the expression of apoptosis-related proteins in each group treated with Jasmine. The effects of pcDNA-CAV-1 and Jasmine on the protein expression levels of Occludin and Claudin-1 were detected by Western Blot method. Results The knockdown efficiency of CAV-1-siRNA and the overexpression efficiency of pcDNA-CAV-1 were verified in HT-29 cells. Jasmine can promote the proliferation level and the expression level of anti-apoptotic proteins of HT-29 cells in the inflammatory bowel disease cell model by up-regulating CAV-1. Jasmine can promote the protein expression levels of Occludin and Claudin-1 by up-regulating CAV-1. Conclusion Jasmine promotes the protein expression levels of Occludin and Claudin-1 by up-regulating CAV-1, thereby promoting the proliferation level and inhibiting the apoptosis level of HT-29 cells, playing a protective role in the inflammatory bowel disease cell model.
