摘要
将20只SPF级妊娠Wister母鼠随机分为常温对照组和冷应激组,每组10只。对照组妊娠母鼠妊娠期全程在(22±2)℃的环境中饲养,冷应激组母鼠在产前7d置于(4±0.1)℃人工智能气候室中饲养,分别冷刺激3d和7d。麻醉后采集各时间点(每个时间点5只妊娠母鼠)肝脏组织样本,常温对照组在相同时间点采集肝脏组织样本。采用Westen blot方法检测肝脏中HSP70、P65蛋白的表达量。结果显示:冷应激组妊娠母鼠肝脏中HSP70和P65蛋白表达量均高于常温对照组,且差异均有统计学意义(P〈0.05)。结果表明:产前冷应激使妊娠母鼠肝脏中HSP70、P65蛋白表达量升高,推测这有可能促使肝脏中炎症反应的发生。
To study the effect of prenatal cold stress on the expression of HSP70 and p65 protein in the liver of pregnant rats. Twenty SPF pregnant rats were randomly divided into normal control group and cold stress group (n= 10). In the control group, the pregnant rats were housed in the environment of (22±2)℃ during the pregnancy. The cold stress group was housed in the artificial intelligence climate chamber at (4±0.1)℃ 7 days before delivery,cold stimulation for 3 days and 7 days respectively. After anesthesia, the liver samples were collected at each time point (5 preg- nant mothers at each time point). The liver samples were collected in the normal temperature con- tro[ group at the same time point. Western blot was used to detect the expression of HSP70 and P65 protein in the liver. The results showed that the expression of HSP70 and P65 protein in the cold stress group were higher than those in the control group (P〈0.05). It is found that prenatal cold stress increases the expression of HSPT0 and P65 protein in the liver of pregnant rats, suggesting that this may lead to the occurrence of inflammatory reaction in the liver.
