强心安神方对慢性心衰大鼠心肌细胞凋亡的保护作用及机制探讨 被引量:1

摘要 目的:探讨强心安神方对心衰大鼠心肌细胞凋亡的影响。方法:Wister大鼠随机分成正常对照组、模型组、强心安神方小、大剂量组及卡托普利组,灌胃给药4周后,TUNEL法检测心肌细胞凋亡指数,心脏超声测量大鼠射血分数(LVEF)及左室收缩率(LVFS),Real Time-PCR法检测内质网应激标志性基因GRP78、CHOP mRNA表达。结果:与模型组比较强心安神方小剂量组、强心安神方大剂量组、卡托普利组心肌排列整齐致密,心肌细胞凋亡减少;与模型组相比,强心安神方小剂量组及大剂量组GRP78 mRNA表达降低;与正常对照组相比,模型组、强心安神方小剂量组、强心安神方大剂量组、卡托普利组CHOPmRNA表达升高,差异均有统计学意义。结论:强心安神方可以改善心力衰竭大鼠心功能,抑制心肌细胞凋亡,其机制可能与调控GRP78 mRNA表达有关。 Objective: To investigate the effect of Qiangxin Anshen recipe on cardiomyocyte apoptosis in rats with heart failure. Methods: Wister rats were randomly divided into normal control group, model group, Qiangxin Anshen Fang small and large dose groups and captopril group. Rat ejection fraction(LVEF) and left ventricular contraction rate(LVFS), Real Time-PCR method to detect endoplasmic reticulum stress marker gene GRP78, CHOP mRNA expression. Results: Compared with the model group, the small-dose Qiangxin Anshen Prescription group, the large-dose Qiangxin Anshen Prescription group, and the captopril group had neatly arranged and compact myocardium, and decreased myocardial cell apoptosis;compared with the model group, the small-dose Qiangxin Anshen Prescription Compared with the normal control group, the expression of CHOPmRNA in the model group, Qiangxin Anshen Prescription low-dose group, Qiangxin Anshen Prescription high-dose group, and captopril group increased, and the differences were statistically significant. learning meaning. Conclusion: Qiangxin Anshen Prescription can improve cardiac function and inhibit cardiomyocyte apoptosis in rats with heart failure, and its mechanism may be related to the regulation of GRP78 mRNA expression.
