摘要
目的:探讨强心安神方对心衰大鼠心肌细胞凋亡的影响。方法:Wister大鼠随机分成正常对照组、模型组、强心安神方小、大剂量组及卡托普利组,灌胃给药4周后,TUNEL法检测心肌细胞凋亡指数,心脏超声测量大鼠射血分数(LVEF)及左室收缩率(LVFS),Real Time-PCR法检测内质网应激标志性基因GRP78、CHOP mRNA表达。结果:与模型组比较强心安神方小剂量组、强心安神方大剂量组、卡托普利组心肌排列整齐致密,心肌细胞凋亡减少;与模型组相比,强心安神方小剂量组及大剂量组GRP78 mRNA表达降低;与正常对照组相比,模型组、强心安神方小剂量组、强心安神方大剂量组、卡托普利组CHOPmRNA表达升高,差异均有统计学意义。结论:强心安神方可以改善心力衰竭大鼠心功能,抑制心肌细胞凋亡,其机制可能与调控GRP78 mRNA表达有关。
Objective: To investigate the effect of Qiangxin Anshen recipe on cardiomyocyte apoptosis in rats with heart failure. Methods: Wister rats were randomly divided into normal control group, model group, Qiangxin Anshen Fang small and large dose groups and captopril group. Conclusion: Qiangxin Anshen Prescription can improve cardiac function and inhibit cardiomyocyte apoptosis in rats with heart failure, and its mechanism may be related to the regulation of GRP78 mRNA expression.
