Study on the Introduction of Insect-resistant Gene into Oilseedrape Using Agrobacterium tumefaciens-mediated Transformation

摘要 以油菜的子叶柄作为转化的受体材料 ,经农杆菌感染和共培养 ,在含卡那霉素的筛选培养基上诱导产生了大量的不定芽 .经生根培养 ,获得了再生植株 .通过PCR扩增和基因组Southern杂交分析 ,证明其中 5株为转基因植株 .对转基因植株进行抗虫性检测 ,有 2株表现出较好的抗虫性 . WT5'BZThe cotyledon petiols of oilseed rape WTBX(Brassica napus)WTBZ were used as recipient of transformation. After being infected and co-cultured with WTBXAgrobacterium tumefaciens,WTBZcotyledon petiols produced a lot of adventitious buds on selection medium supplemeted with kanamycin. Kanamycin-resistant plants were then obtained from the adventitious buds by rooting culture. DNA Southern blot hybridization and polymerase chain reaction showed that 5 of the plants were transgenic when probed with the insect-resistant gene. These transgenic plants exhibited different insect-resistant response when they were subjected to leaf feeding experiments, but two of them displayed a good resistance to pest insect larvae.
