摘要
通过土壤农杆菌(Agrobacterium tumefaciens)的介导,将烟草花叶病毒(TMV)的54×103蛋白cDNA转入番茄植物中,得到了转基因植株.这些植株抗卡那霉素,具有胭脂碱合成酶的活性.DNA的PCR扩增及Northern blot分析表明,54×103蛋白基因已整合到番茄基因组上.攻毒实验表明,未转基因的对照组接种病毒2周后即出现花叶,并且随着时间的推移,花叶症状更加典型.而转基因的植物接种病毒后一直到开花结果都未见发病,即没有花叶出现.
The cDNA encoding 54 x 103 protein of tobacco mosaic virus(TMV)is introducedinto tomato by Agrobacterium-mediated method and transgenic plants are obtained.Nopalinesynthtase activity is demonstrated in the kanamycin-resistance lantlet.It is demonstrated thatTMV 54 x 103 protein gene has been integrated into the genomes of tomato plants by PCR andNorthern blot. The transgenic tomatoes show no typical mosaic symptom after TMV infection inthe Whole life while the control plants show typical mosaic synptoms in 2 weeks after TMVinfection.
