鸭油甘油二酯对肥胖大鼠减肥降脂作用与机制研究 被引量:3

Effects and Mechanism of Duck Oil Diglyceride on Weight Loss and Lipid Reduction of Obese Rats

摘要 本试验旨在研究鸭油、鸭油甘油二酯对肥胖大鼠体重、器官指数、血清生化指标的影响,以及鸭油甘油二酯抗肥胖和降胆固醇作用的潜在机制。试验分为2个阶段。造模阶段:将80只大鼠随机分为2个组,对照组每组4个重复,每个重复5只;造模组每组12个重复,每个重复5只。对照组大鼠自由采食基础饲粮,造模组大鼠自由采食脂肪含量为60%的高脂饲粮,连续饲养28 d。修复干预阶段:将造模组的肥胖大鼠随机分为模型对照组、鸭油组、鸭油甘油二酯组,每组4个重复,每个重复5只。模型对照组以蒸馏水灌胃,用高脂饲粮连续喂养30 d,其余各组分别用鸭油、鸭油甘油二酯进行灌胃,用基础饲粮连续喂养30 d。结果表明:1)与对照组相比,造模组的体重显著升高(P<0.05),表明造模成功。2)与模型对照组相比,鸭油甘油二酯组的最终体重、Lee’s指数、肝体比以及血清甘油三酯、总胆固醇、低密度脂蛋白胆固醇含量均显著降低(P<0.05);血清高密度脂蛋白胆固醇含量升高,但差异不显著(P>0.05);心体比、脾体比、肾体比、睾体比均无显著差异(P>0.05)。3)与模型对照组相比,鸭油甘油二酯组的肝脏甘油三酯、磷脂酰胆碱(PC)、胆固醇脂(CE)、甘油二酯、酰基肉碱(CAR)、溶血磷脂酰胆碱(LPC)和游离脂肪酸(FFA)含量显著降低(P<0.05)。鸭油甘油二酯能够影响脂肪细胞的甘油酯代谢、产热作用和脂肪分解调节途径、磷脂酶D的信号通路、胆固醇代谢等相关代谢途径。由此可见,鸭油甘油二酯具有减肥降脂的功效,而调节脂质稳态可能是其发挥减肥降脂作用机制的关键途径。 This experiment was conducted to investigate the effects of duck oil and duck oil diglyceride on body weight,organ indexes and serum biochemical indexes of obese rats,and the potential mechanism of anti-obesity and cholesterol-lowering effects of duck oil diglyceride. The experiment was divided into two stages. Modeling stage:eighty rats were randomly divided into 2 groups,there were 4 replicates per group and 5 rats per replicate in the control group,and there were 12 replicates in each group and 5 replicates in each replicate in the modeling group. Rats in the control group were freely fed a basal diet,and rats in the modeling group were freely fed a high fat diet containing 60% fat,continuous feeding for 28 days. Repair intervention stage:the obese rats in the modeling group were randomly divided into model control group,duck oil group and duck oil diglyceride group,with 4 replicates in each group and 5 rats in each replicate. Rats in the model control group were gavaged with distilled water and fed a high fat diet for 30 days,and rats in other groups were gavaged with duck oil and duck oil diglyceride,respectively,and fed basal diets for 30 days. The results showed as follows:1)compared with the control group,the body weight of the modeling group was significantly increased(P<0.05),indicating that the modeling was successful. 2)Compared with the model control group,the final body weight,Lee’s index,liver body ratio and contents of triglyceride,total cholesterol and low density lipoprotein cholesterol in serum of duck oil diglyceride group were significantly decreased(P<0.05);the serum high density lipoprotein cholesterol was increased,but no significant difference(P > 0. 05);the heart body ratio,spleen body ratio,kidney body ratio and testis body ratio were no significant difference(P>0.05). 3)Compared with the model control group,the contents of triglyceride,phosphatidylcholine(PC),cholesterol ester(CE),diglyceride,acylcarnitine(CAR),lysophosphatidylcholine(LPC)and free fatty acid(FFA)in liver of duck oil diglyceride group were significantly decreased(P<0.05). The duck oil diglyceride could affect the adipocyte metabolism,thermogenesis,adipocalysis regulation,phospholipase D signaling pathway,cholesterol metabolism and other related metabolic pathways. In conclusion,duck oil diglyceride has the effect of weight loss and lipid reduction,and regulating lipid homeostasis may be the key mechanism of weight loss and lipid reduction.[Chinese Journal of Animal Nutrition,2022,34(6):4020-4031]
