摘要
目的观察人表皮生长因子受体2(HER2)、沉默信息调节因子1(SIRT1)及人第10号染色体缺失的磷酸酶与张力蛋白同源物基因(PTEN)在膀胱尿路上皮细胞癌中的表达及对预后的影响。方法选择2018年2月—2019年3月收治的124例膀胱尿路上皮细胞癌的癌组织标本设为研究组,并选取距癌组织3 cm处的正常组织作为对照组。比较两组HER2、SIRT1及PTEN表达情况,同时根据预后情况分组,分析影响膀胱尿路上皮细胞癌患者预后的危险因素。结果研究组HER2、SIRT1的阳性表达率高于对照组,PTEN阳性表达率低于对照组(P<0.05)。多因素Logistic回归分析显示,有家族史、WHO病理分级Ⅱ~Ⅲ级、UICC-TNM临床分期T_(2)~T_(4)期、HER2表达阳性、SIRT1表达阳性及PTEN表达阴性均为影响膀胱尿路上皮细胞癌患者预后的独立危险因素(P<0.05,P<0.01)。结论HER2、SIRT1在膀胱尿路上皮细胞癌组织中呈高表达,PTEN呈低表达,均为影响膀胱尿路上皮细胞癌患者预后的独立危险因素。
Objective To observe expressions of human epidermal growth factor receptor 2(HER2),silent mating type information regulation 2 homolog1(SIRT1)and gene of phosphate and tension homology deleted on chromosome ten(PTEN)in patients with bladder urothelial carcinoma(BUC)and their effects on prognosis.Methods Cancer tissue specimens from 124 patients with BUC who had admitted between February 2018 and March 2019 were selected as research group,and normal tissue 3 cm away from the cancer tissues were selected as control group.Expressions of HER2,SIRT1 and PTEN were compared between two groups,and they were grouped simultaneouly according to the prognosis to analyze risk factors affecting prognoses of patients with BUC.Results Compared with those in control group,positive expression rates of HER2 and SIRT1 were significantly higher,while positive expression rate of PTEN was significantly lower in research group(P<0.05).Multivariate Logistic regression analysis showed that family history,world health organization(WHO)pathological grade Ⅱ-Ⅲ,international union against cancer-tumour node metastases(UICC-TNM)clinical stage T_(2)-T_(4),positive expression of HER2,positive expression of SIRT1 and negative expression of PTEN were all independent risk factors affecting prognoses of patients with BUC(P<0.05,P<0.01).Conclusion Highly expressed HER2 and SIRT1 and lowly expressed PTEN in BUC tissues are independent risk factors affecting prognoses of patients with BUC.
