摘要
目的研究人参蜜百部醇提物对烟熏法诱导慢性支气管炎模型大鼠的影响。方法将大鼠随机分为模型组、空白组、阳性药组、人参蜜百部高、中、低剂量组,每组10只,共计60只。除空白组外,各组大鼠放入烟熏仓内,每天烟熏两次,进行造模,每次30min,连续6周。第4周后开始灌胃给药,阳性药组给予醋酸地塞米松1 mg/kg,人参蜜百部高剂量组12 g/kg,人参蜜百部中剂量组6 g/kg,人参蜜百部低剂量组3 g/kg,空白组、模型组给予等量蒸馏水。末次给药30min后,记录大鼠在5min内的咳喘次数,测定大鼠血清中SOD(超氧化物歧化酶)、MDA(丙二醛)、TNF-α(肿瘤坏死因子)、IL-6(白介素-6)、IL-1β(白细胞介素-1)。结果与空白组比较,模型组大鼠咳喘次数明显增加(P<0.01),血清中SOD含量明显下降(P<0.01)、MDA含量明显升高(P<0.01),模型组大鼠TNF-α、IL-6.IL-1β含量明显增加(P<0.01);给药后,与模型组相比,醋酸地塞米松组、人参蜜百部高冲剂量组大鼠咳喘次数均显著降低(P<0.01),人参蜜百部低剂量组(P<0.05),各给药组大鼠血清SOD明显升高(P<0.01)、MDA明显下降(P<0.01),醋酸地塞米松组、人参蜜百部高、中剂量组大鼠血清中TNF-α、IL-6、IL-1β含量均显著降低(P<0.01),人参蜜百部低剂量组TNF-α含量显著降低(P<0.05)。结论人参蜜百部醇提物可明显缓解大鼠因烟熏法诱导的慢性支气管炎,其作用可能与调节SOD、MDA、TNF-α、IL-6、IL-1β等因子的水平有关。
Objective To observe the effect of ethanol extract of Ginseng and processed Stemonae on smoke-induced chronic bronchitis rats.Methods Sixty healthy male SD rats were randomly divided into blank group,model group,dexamethasone acetate group,Ginseng and processed Stemonae high-dose group,medium dose group and low-dose group.Except the blank group,the rats in each group were smoked in the self-made smoke warehouse for 6 weeks,twice a day for 30 minutes each time.After 4 weeks of smoking,the positive drug group was given dexamethasone acetate(1mg/kg),the high-dose group(12g/kg),the middle dose group(6g/kg),and the low-dose group(3g/kg).The blank group and modol group was given the same amount of distilled water.The changes of the times of cough and asthma in 5 min were recorded,and the serum levels of SOD(superoxide dismutase),malondialdehyde(MDA),TNF-α(tumor necrosis factor),IL-6(interleukin-6)and IL-1β(interleukin-1)were measured.Results Compared with the blank group,the number of cough and asthma in model group was significantly increased(P<0.01),the content of SOD in serum of model group was significantly decreased(P<0.01),MDA was significantly increased(P<0.01),TNF-α,IL-6,IL-1β in model group were significantly increased(P<0.01);after administration,compared with model group,the number of cough and asthma of rats indexamethasone acetate group and Ginseng and processed Stemonae high and medium group was significantly decreased(P<0.01),while that in the low-dose group(P<0.05),the serum SOD and MDA levels in each treatment group were significantly increased(P<0.01),and the contents of TNF-α,IL-6 and IL-10 in dexamethasone acetate group,high and medium dose groups of Ginseng and processed Stemonae were significantly decreased(P<0.01),and the contents of TNF-α,IL-6 and IL-1β were significantly decreased(P<0.01)Conclusion The ethanol extract of Ginseng and processed Stemonae can significantly alleviate chronic bronchitis induced by smoking in rats,and the reason for its action may be to regulate the levels of SOD,MDA,TNF-α,IL-6,IL-1β and other factors.
