摘要 【目的】研究茉莉酸(JA)生物合成抑制剂对马铃薯离体块茎形成和发育的影响,为揭示JA对马铃薯块茎发育的调控机理提供理论依据.【方法】以马铃薯试管苗诱导的匍匐茎为试验材料,外源添加不同浓度JA生物合成抑制剂水杨苷羟肟酸(SHAM),培养40 d后匍匐茎诱导形成离体块茎,分析匍匐茎结薯率以及块茎直径、鲜干质量、淀粉含量、葡萄糖含量、果糖含量、蔗糖含量和脂氧合酶(LOX)活性变化.【结果】在一定浓度范围内,随着SHAM浓度增加,匍匐茎结薯率、块茎直径、鲜干重和LOX活性呈现逐渐下降趋势,淀粉含量先增加后降低,葡萄糖、果糖和蔗糖含量均先降低后增加.当SHAM浓度达到2400μmol/L时,匍匐茎不再膨大形成块茎,同时外源施加5μmol/L JA,仍无块茎形成.【结论】外源添加JA生物合成抑制剂会阻碍内源JA合成,不利于块茎形成,说明JA在马铃薯块茎形成和发育过程中起重要调控作用. 【Objective】The effect of jasmonic acid(JA)synthesis inhibitor on potato tuber formation and development was investigated,which provides a theoretical basis for revealing the regulation mechanism of JA on potato tuberization.【Method】The potato stolon in vitro was used as materials,and the different concentrations of JA synthesis inhibitor,salicinhydroxamic acid(SHAM),were added in tuber-induction medium.After 40 d cultures,the tubers under each treatment were obtained.The percentage of tuberization,tuber diameter,tuber fresh and dry weight,sugars content(glucose,fructose and sucrose)and starch content of tubers,lipoxygenase(LOX)activity of tubers were analyzed under different SHAM concentrations treatment.【Result】With the increase of SHAM concentration,the percentage of tuberization,tuber diameter,tuber fresh and dry weight,and LOX activity of tubers were gradually decreased.The starch content of tubers was firstly increased and then decreased,whereas the content of glucose,fructose and sucrose were firstly decreased and then increased.When the concentration of SHAM was 2400μmol/L,the stolon no longer expands to form tubers.Even if exogenously adding 5μmol/L JA under 2400μmol/L SHAM treatment,tubers still cannot be formed.【Conclusion】Exogenous JA biosynthesis inhibitors inhibited endogenous JA synthesis,which was not conducive to tuber formation.It is indicated that JA plays an important regulatory role in potato tuber formation and development.
