摘要
通过分析 4种内源激素IAA、GA3 、ABA和JA在试管地黄形态建成中的动态变化 ,初步探讨了试管地黄形成的生理生化机制。在MS +6 BA 0 5mg·L-1+NAA 0 1mg·L-1+5 %蔗糖培养基中诱导地黄无菌苗茎段形成不定根 ,再转入MS +6 BA 1 0mg·L-1+IBA 0 1mg·L-1+5 %蔗糖继代培养 ,取不同生长时期的根 ,对其内源激素进行分离和纯化 ,经HPLC法进行测定 ,结果表明在试管地黄形成过程中IAA、GA3 、ABA和JA的含量存在动态变化 ,基本呈上升趋势。
Through analysis of variation and function of four endogenous hormones (IAA,GA 3, ABA and JA) in the formation of tuberous roots of Rehmannia glutinosa in vitro to explore the physiological and biochemical mechanism of tuberous root development. The endogenous hormones of tuberous roots were isolated in different stages of root expansion when they were cultured on MS+6-BA 1.0 mg·L -1+IBA 0.1 mg·L -1+sucrose 5% medium, then purified and detected with High Performance Liquid Chromatography System. The results showed that the endogenous hormones varied with tuberous root development and thus played an important role in controlling the root formation.
