肾病综合征鼠模型生长障碍的特点及机制 被引量:2

CHARACTERISTIC AND PATHOGENESIS OF GROWTH FAILURE IN RATS WITH NEPHROTIC SYNDROME

摘要 目的 :探讨肾病综合征 (NS)鼠生长障碍的特点及机制。方法 :制备大鼠阿霉素NS模型 ,测NS鼠和正常鼠的身长、体重、尿蛋白、血清生化指标 ,用放免法和免疫放射法分别测定血、尿IGF -I、IGFBP - 3水平 ,用RT -PCR法测定肝组织IGF -ImRNA、IGFBP - 3mRNA表达水平。结果 :第 4周末 ,NS组进食量、体重增长和身长增长均低于正常组 ,P <0 .0 5 ;NS组 2 4hr尿蛋白定量高于正常组 ,P <0 .0 5 ;NS组血Alb、Chol、TG水平低于正常组 ,P <0 .0 5 ;NS组血IGF -I、IGFBP - 3水平低于正常组 ,而尿IGF -I、IGFBP - 3高于正常组 ,P <0 .0 5 ;两组肝IGF -ImRNA表达量之间的差别无统计学意义 (P >0 .0 5 ) ;NS组肝IGFBP - 3mRNA表达量低于正常组 ,P <0 .0 1。相关分析 :NS组的血IGF -I与尿IGF -I负相关 ,血IGFBP水平与身长正相关 ,肝IGFBP - 3mRNA表达量与血IGFBP水平正相关。结论 :NS鼠存在生长障碍和IGF -I、IGFBP - 3代谢紊乱 ;血IGF -I、IGFBP - 3浓度降低是NS鼠生长障碍的主要原因之一。 Objective:To study the Characteristic and pathogenesis of growth failure in rats with nephrotic syndrome(NS).Methods:Preparing NS rats model with adriamycin,serum and urine concentrations of IGF-I and IGFBP-3 were assayed by RIA and IRMA respectively; and IGF-I mRNA and IGFBP-3 mRNA levels in liver were assayed by RT-PCR.Weight,urinary protein,serum biochemital index and length were measured in every week.Results:Weight gain and length gain, serum Alb,Chol and TG, serum IGF-I and IGFBP-3 levels in NS group were significantly lower than that of the control group by the end of the fourth week( P <0.05). Quantitative analysis of 24 hours urinarly protein, urine IGF-I and IGFBP-3 levels were higher than that of the control group ( P <0.05). Liver IGF-I mRNA expression levels in NS group were similar to that of the control rats, but hepatic IGFBP-3mRNA levels in NS group were reduced( P <0.01). There was a significantly negative correlation between serum IGF-I and urine IGF-I in NS group.There was a significantly positive correlation between serum IGFBP-3 and height, and so did between liver IGFBP-3 mRNA levels and serum IGFBP-3. Conclusion:Abnormal IGF-I and IGFBP-3 metabolism and growth failure were observed in nephrotic rats .The low serum IGF-I levels were due to increased urinary loss of IGF-I, and the low serum IGFBP-3 levels were due to both decreased synthesis and increased urinary loss. The low serum IGF-I and IGFBP-3 levels may contribute to growth failure in nephrotic rats.
