摘要
目的:探讨补中益气汤对急性脑缺血大鼠行为学及脑损伤的影响。方法:选取50只成年雄性SD大鼠按随机数字表法分为假手术组、模型对照组、低、中、高剂量组,除假手术组外均建模。3剂量组分别给予补中益气汤0.5、1.0、1.5 g/kg灌胃,假手术组和模型对照组均给予2 ml生理盐水灌胃,持续2周。比较各组行为学、脑指数并观察病理学变化,检测对比海马区组织凋亡相关蛋白的表达情况。结果:与假手术组比较,模型对照组BBB评分、Rivlin-Tator斜板试验评价结果、水迷宫实验、脑指数均显著较差,海马区组织病理变化严重,且Caspase-3、Bax蛋白相对表达量显著升高,而Bcl-2蛋白表达量显著降低。经过治疗后3剂量组上述指标均有改善,其中高剂量组效果最为显著。结论:补中益气汤能改善急性脑缺血大鼠的行为学,减轻脑损伤,推测与上调Bcl-2蛋白表达、下调Caspase-3和Bax蛋白表达有关。
Objective:To explore the effect of Buzhong Yiqi decoction on behavior and brain injury in rats with acute cerebral ischemia.Methods:50 adult male SD rats were randomly divided into sham operation group,model control group,low,medium and high dose group,except the sham operation group,the other groups were all set up models.The 3 dose groups were given 0.5,1.0,1.5 g/kg Buzhong Yiqi solution,and the sham operation group and the model control group were given 2ml physiological saline for 2 weeks.The praxiology and the brain indexes were compared,and the pathological changes were observed,and the expressions of hippocampus tissues related protein were detected in the brain tissues.Results:Compared with sham operation group,BBB score,Rivlin-Tator slant test result,Morris water maze and brain index of model control group were significantly worse,of which the pathological changes of brain tissue were serious,and the relative expressions of Caspase-3 and Bax increased significantly,while the expression level of Bcl-2 protein was significantly decreased.After treatment,all the above indexes were improved in the 3 dose groups,and the effect of the high dose group was the most significant.Conclusion:Buzhong Yiqi decoction can improve the praxiology of acute cerebral ischemia rats,reduce brain injury.It is presumed that it is related to the up regulation of the expression of Bcl-2 protein,the down regulation of Caspase-3 and the expression of Bax protein.
