摘要 目的 分析胰腺癌中bcl-2基因的mRNA转录和蛋白翻译,探讨其在胰腺癌发生、发展中的作用。方法 以50例胰腺癌组织、15例胰腺癌转移灶组织和10例慢性胰腺炎为研究对象,采用免疫组织化学(SP法)和原位分子杂交法检测bcl-2的表达。结果 Bcl-2蛋白表达与患者年龄、性别、TNM分期无关,与肿瘤分化程度、有无转移病灶有关。在胰腺癌组织中的表达低于胰腺炎组织,但明显高于转移灶。免疫组织化学与原位杂交技术检测胰腺癌组织中的bcl-2基因表达结果一致。结论 Bcl-2蛋白可能作用于胰腺细胞的癌变阶段,并与胰腺导管上皮癌变有关;bcl-2基因在mRNA和蛋白水平表达是相对稳定的。 Objective To investigate mRNA transcription and protein translation of bcl-2 gene and the role of bcl-2 gene in the development and progression of pancreatic cancer. Methods Expression of Bcl-2 and bcl-2 mRNA was detected immunohistochemically by mRNA and in situ molecular hybridization techniques. In 50 cases of pancreatic cancer tissues and in 15 cases of metastatic pancreatic cancer tissues. 10 cases of chronic pancreatitis tissues were used as control group. Results Expression of Bcl-2 protein was not correlated with age, gender and TNM classification of the patients, but with differentiation of the tumors and with the presence or absence of metastasis. Expression of Bcl-2 protein in pancreatic cancer group was lower than that in pancreatitis group, but significantly higher than that in the metastatic group. In pancreatic cancer, the result of bcl-2 gene expression detected by the immunohistochemical technique was consistent with that detected by the in situ molecular hybridization technique. Conclusions Bcl-2 protein may play a role in the canceration course of pancreas cells and be related with epithelial canceration of the pancreas duct. Expression of bcl-2 gene is relatively stable at mRNA and protein level.
