摘要
目的:探讨自噬相关蛋白在卡培他滨联合奥沙利铂治疗晚期乳腺癌中的表达及其主要作用机制,并分析卡培他滨联合奥沙利铂对乳腺癌患者抗肿瘤免疫功能的影响。方法:选取本院2015年1月~2017年1月收治的60例女性原发性乳腺癌晚期患者,接受卡培他滨联合奥沙利铂化疗2疗程后评价疗效。采用免疫组织化学法检测乳腺癌患者癌组织中m TOR和LC3-Ⅱ蛋白的表达;采用Real-Time PCR法和Western blot法检测乳腺癌组织中m TOR、LC3-II、Beclin-1、p62和Atg5mRNA和蛋白的表达;流式细胞仪检测患者在卡培他滨联合奥沙利铂治疗前后CD3^+、CD4^+、CD8^+、CD4^+/CD8^+及CD16^+/CD56^+水平。结果:卡培他滨联合奥沙利铂化疗总有效率56.67%。卡培他滨联合奥沙利铂化疗后乳腺癌血清中CD3^+、CD4^+、CD4^+/CD8^+、CD16^+/CD56^+表达量显著升高,CD8^+含量显著降低,差异有统计学意义(P<0.05)。与化疗前乳腺癌组织相比,卡培他滨联合奥沙利铂化疗后乳腺癌组织中m TOR和p62 mRNA表达和蛋白含量显著降低,LC3-II、Beclin-1和Atg5 mRNA表达和蛋白含量显著上升,差异有统计学意义(P<0.05)。结论:卡培他滨联合奥沙利铂对晚期乳腺癌具有较好的疗效,且自噬相关蛋白介导的m TOR信号通路在此过程中发挥重要的作用。
Objective:To investigate the expression of autophagy-related protein in capecitabine and oxaliplatin in the treatment of advanced breast cancer and its main mechanism,and to analyze the changes of anti-tumor immune function in patients with breast cancer before and after treatment.Methods:Sixty patients with advanced primary breast cancer who were treated in our hospital from January 2015 to January 2017 were enrolled in this study.After 2 cycles of capecitabine and oxaliplatin chemotherapy.Immunohistochemistry was used to detect the expression of mTOR and LC3-II protein in breast cancer tissues.The mRNA and protein expressions of mTOR,LC3-II,Beclin-1,p62 and Atg5 in breast cancer tissues were detected by Real-Time PCR and Western Blot;Flow cytometry was used to detect CD3^+,CD4^+,CD8^+,CD4^+/CD8^+and CD16^+/CD56^+levels in patients with capecitabine and oxaliplatin before and after treatment.Results:Capecitabine combined with oxaliplatin chemotherapy the total effective rate 56.67%.The levels of CD3^+,CD4^+,CD4^+/CD8^+,CD16^+/CD56^+in breast cancer serum after oxcarbazepine plus capecitabine combined with oxaliplatin were significantly increased(P〈0. 05).Compared with pre-chemotherapy breast cancer tissue,the expressions of m TOR and p62mRNA and protein were significantly decreased in breast cancer tissues after the combination of capecitabine and oxaliplatin chemotherapy.The expressions of LC3-II,Beclin-1 and Atg5 mRNA and protein were significantly Rise,with significant statistical difference(P〈0. 05).Conclusion:Capecitabine combined with oxaliplatin has a good therapeutic effect on advanced breast cancer,and the autophagy-related protein-mediated m TOR signaling pathway plays an important role in this process.
