Simultaneous determination of four constitutents in Baishi Wan by HPLC gradient elution with switching wavelength

摘要 目的建立高效液相-梯度洗脱-波长切换法同时测定白蚀丸中二苯乙烯苷、补骨脂素、异补骨脂素和丹皮酚的质量分数。方法采用Dikma Spursil色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈-水,梯度洗脱;流速:1.0 mL/min;柱温:30℃;检测波长:320 nm(二苯乙烯苷),246 nm(补骨脂素和异补骨脂素)和274 nm(丹皮酚);进样量:5μL。结果白蚀丸中的二苯乙烯苷、补骨脂素、异补骨脂素和丹皮酚分别在6.26~43.84μg/mL(r=0.999 8)、3.99~23.93μg/mL(r=0.999 4)、2.40~33.57μg/mL(r=0.999 3)、0.80~4.00μg/mL(r=0.999 9)范围内与峰面积呈良好的线性关系;平均回收率分别为82.8%(RSD=1.6%)、101.4%(RSD=1.8%)、102.1%(RSD=1.6%)、103.0%(RSD=1.9%)。结论所建立的方法可为控制白蚀丸的质量提供依据。 Objective To develop an HPLC-DAD gradient elution with switching wavelength for simultaneous determination of stilbeneglycoside,psoralen,isopsoralen and paeonol in Baishi Wan. Methods The analysis was performed on a Dikma Spursil C18 column with its temperature maintained at 30 ℃,mobile phase comprised of water and acetonitrile with gradient elution at the flow rate of 1.0 mL/min,and detection wavelengths of 320 nm for stilbeneglycoside,246 nm for psoralen and isopsoralen,and 274 nm for paeonol.Results The stilbeneglycoside,psoralen,isopsoralen and paeonol in the Bai Shi Wan showed a good linear relationship with peak area in the range of 6. 26-43. 84 μg/mL( r = 0. 999 8), 3. 99-23. 93 μg/mL( r = 0.999 4),2.40-33.57 μg/mL( r = 0.999 3),and 0.80-4.00 μg/mL( r = 0.999 9),respectively. The average recovery rates were 82. 8%( RSD = 1. 6%), 101. 4%( RSD = 1. 8%), 102. 1%( RSD = 1. 6%),103.0%( RSD = 1. 9%),respectively. Conclusion The established method was validated and suitable for the quality control of Baishi Wan.
