超高效液相色谱法测定贵州烟草中的游离氨基酸 被引量:12

摘要 游离氨基酸在烟草质量控制及卷烟配方优化中具有重要的作用。本研究采用柱前衍生-超高效液相色谱法对贵州烟草中游离氨基酸进行分离测定。结果:该方法可以在8.3 min内实现19种游离氨基酸的分离与测定,其回收率为83.19%~106.98%,相对标准偏差在0.53%~7.39%,检出限(S/N=3)在0.01~0.74μg·mL^(-1),线性相关系数均大于0.999。对贵州省5个产地烟叶样品批量检测,结果:毕节地区与其他地区烟叶样品中游离氨基酸总量差异显著;5个地区烟叶样品中精氨酸、天冬酰胺、谷氨酸、甘氨酸、组氨酸、赖氨酸、苯丙氨酸及脯氨酸含量差异显著,且19种游离氨基酸中脯氨酸含量最高,占总氨基酸含量的70%至80%。 Free amino acids play an important role in tobacco quality control and cigarette formulation optimization. In this study,the separation and determination of free amino acids in Guizhou Tobacco were carried out,using Ultra-high Performance liquid chroma-tography ( UPLC ) with pre-column derivatization. The results showed that the separation of 19 amino acids in 8. 3 min was achieved. The recoveries of the 19 amino acids were in the range of 83. 19% ·106. 98% with the relative standard deviation of 1. 8% · 3. 9%,and the detection limits were 0. 01 ·0. 74pg · mL^-1 Each amino acid showed a good linear relationship with the correlation coefficients more than 0. 999. Batch detection of tobacco samples from five major tobacco-producing regions of Guizhou province were performed. The results showed that there were significant differences in the total contents of free amino acids in tobacco sam-ples from Bijie and other areas. The contents of free arginine, asparagine, glutamic acid, glycine, histidine, phenylalanine and proline were significantly different, and the content of free proline was the highest among the 19 free amino acids, accounted for 70% to 80% of the total content of free amino acids.
