摘要
目的:建立2,4-二硝基氟苯柱前衍生化法测定复方氨基酸注射液(18AA)含量的HPLC方法。方法:以正交回归试验, 考察色谱条件(柱温27-29℃、流动相缓冲液的pH 8.2-9.2和磷酸盐浓度0.008-0.014 mol·L-1)对各氨基酸分离度的影响。结果:最佳色谱条件:柱温28℃,流动相缓冲液pH 8.2和磷酸盐浓度0.014 mol·L-1。结论:本法能很好地分离18种复方氨基酸,含量测定准确,重复性好。
Objective:To separate and determine the compound amino acids injection(18AA) and establish the determination method. Method:The orthogonal test - pluralistic regression method was used and studying the effects of column oven temperature 27-29℃,pH 8. 2 -9. 2 and phosphate concentration 0. 008 -0. 014 mol·L-1 in mobile phase. Results:The best HPLC condition:column oven temperature 28℃,pH 8. 2 and phosphate concentration 0.014 mol·L-1 in mobile phase. Conclusion: The method is accurate with good reproducibility and can be used for compound amino acids(18AA) determination.
