摘要
ZDC-01菌株为分离自海洋、对保护地蔬菜土传病害具有较好防治效果的地下芽孢杆菌(Bacillus subterraneus)。为了提高菌株ZDC-01的活菌数,该研究通过单因子水平筛选和正交试验法对活性菌株ZDC-01的发酵培养基配方和摇瓶发酵条件进行了优化。结果表明,菌株ZDC-01的最佳培养基组成为可溶性淀粉2.0%,豆饼粉3.0%,葡萄糖0.5%,鱼粉0.5%,K_2HPO_4 0.03%,CaCO_3 0.5%,MgSO_4·7H_2O 0.03%,(NH_4)_2SO_40.1%,NaCl 0.3%。最适发酵条件为培养温度30℃、摇床转速200 r/min、发酵起始pH 7.0、接菌量3%、装液量50m L/250m L、发酵时间48h。经验证,优化后发酵菌液活菌数为38×10~9cfu/m L,与初始发酵工艺的发酵菌液含菌量(23×10~9cfu/m L)相比,提高了65.22%。
Bacillus subterraneus ZDC-01,originally isolated from the marine,showed strong biocontrol activitiesagainst a large number of soil-borne diseases of protected vegetable.This paper reports the fermentation medium andthe culture conditions of strain ZDC-01 optimized by single factor horizontal and orthogonal experiments.The resultsshowed that the best medium were Soluble starch 2.0%,soybean cake powder 3.0%,glucose 0.5%,fish meal 0.5%,K_2HPO_4 0.03%,CaCO_3 0.5%,MgSO_4.7 H_2O 0.03%,(NH_4)_2SO_4 0.1%,NaCl 0.3%.The optimal fermentation conditionswere:culture temperature 30℃,rotation speed 200 r/min,initial p H value 7.0,inoculation 3%,liquid volume of 50 m L/250 m L,and fermentation for 48 h.Optimized biological fermentation process resulted in fermentation efficien-cy 38×10~9 cfu/m L,increased by 65.22% in comparison with the initial fermentation process(23×10~9 cfu/m L).
