高效液相色谱法测定蓝莓叶中绿原酸的含量 被引量:3

摘要 采用高效液相色谱法测定蓝莓叶中绿原酸的含量。所用色谱柱为Eclipse XDB C18(250 mm×4.6 mm,2.5μm),流动相为乙腈-0.1%磷酸(体积比为13:87),检测波长327 nm。当绿原酸质量浓度为6~96μg/m L时,以绿原酸对照品溶液的峰面积比为纵坐标(Y)、绿原酸的质量浓度为横坐标(X)进行线性回归,二者呈良好的线性关系,线性方程为:Y=74.15X-432.3,R2=0.999,平均回收率为99.98%,RSD=0.506%(n=6)。按照以上色谱方法检测了3种蓝莓叶中绿原酸的含量,并与蓝莓果实中绿原酸的含量比较,结果显示,3种蓝莓叶中的绿原酸含量显著高于对应品种果实中的绿原酸含量(P<0.05)。 An HPLC method for the quantitative determination of chlorlgenic acid in blueberry leaf was investigated. The operating condi- tions were Eclipse XDB C18 column (250 mm×4.6 mm,2.5μm), mobile phase of acetonitrile-0. 1% H3PO4 (13: 87, V/V), at 1mL/min and UV detection at 327 nm.There was a good linear relationship between the response and the concentration of sampleat the range of 6 - 96mg/ L for chlorlgenic acid. The corresponding regression equation was Y =74. 15X -432. 3, R2 =0. 999. The average recovery was 99. 98% and RSD was 0. 506% (n = 6) . The results showed that this method was simple, specific and accurate. According to the above chromatographic method to detect content of chlorlgenic acid in three kinds of blueberry, and compared with the content of chlorlgenic acid in blueberry fruit, the results showed that three kinds of blueberry leaves the chlorlgenic acid content was higher than the content of chlorlgenic acid in fruit.
