摘要
目的提取枸杞多糖(LBP)、黄芪多糖(APS)、红枣多糖(ZJP),混合制成复合多糖(PM),研究3种多糖及复合多糖的抗氧化作用。方法进行DPPH自由基清除、超氧阴离子自由基清除、羟基自由基清除能力检测。建立H_2O_2氧化损伤的人正常肝细胞(L-O2)和人肝癌细胞(Hep G2)模型,多糖与损伤细胞共孵育后使用MTT法检测细胞活力,评价多糖对细胞抗氧化损伤活性。H_2O_2氧化损伤L-O2和Hep G2细胞后检测丙二醛(MDA)含量,超氧化物歧化酶(SOD)活性和谷胱甘肽过氧化酶(GSH-Px)活性。结果 4种多糖均具有一定的抗氧化活性,但弱于相同浓度的抗坏血酸(VC)。在H_2O_2氧化损伤的细胞模型中,复合多糖显示了在较低浓度即具有良好的保护作用,在质量浓度分别为80μg/m L和160μg/m L时,L-O2细胞和Hep G2细胞存活率最高。4种多糖均能显著降低氧化损伤细胞的MDA水平,细胞SOD和GSH-Px活力均显著升高。结论枸杞多糖、黄芪多糖、红枣多糖及其复合多糖均具有一定的抗氧化作用,较低浓度的复合多糖对氧化损伤的细胞有更好的保护作用。
Objective To study the antioxidant activity of polysaccharides from Lycium barbarum( LBP),Astragalus( APS),Ziziphus jujube( ZJP) and their compound polysaccharide( PM). Methods Antioxidant activity is evaluated by scavenging DPPH free radical, hydroxyl free radical and superoxide anion free radical. L-O2 cells( human normal liver cells) and Hep G2 cells( human liver cancer cells) were damaged by H_2O_2,then the antioxidant activity of polysaccharides in L-O2 cells and Hep G2 cells was detected by MTT assay. The levels of MDA,SOD and GSH-Px were detected. Results Four kinds of polysaccharides showed the antioxidant activity but lower than VC in vitro. The cell viability was the highest in L-O2 and Hep G2 cells when treated with the compound polysaccharide at 80 μg / m L and 160 μg / m L,respectively.Four polysaccharides reduced MDA content and increased SOD and GSH-Px activities in cells. Conclusion LBP,APS,ZJP and PM show antioxidant activity,especially PM at the lower concentration,in cells when damaged by H_2O_2.
