摘要
目的 探究鹿茸提取物对A30P线虫干预效果及其药理作用。方法 购买A30P线虫,按照鹿茸提取物不同浓度分为0、0.4、1、2.5、5 mg/mL组,分析不同浓度下鹿茸提取物对A30P线虫生存率、生长发育、生殖能力、抗氧化能力,并且通过分析同浓度下鹿茸提取物对A30P线虫运动行为、树突恢复情况来评价鹿茸提取物的药理作用。结果 与0 mg/mL组相比,1.0 mg/mL组、2.5 mg/mL组线虫平均寿命以及死亡率无差异(P>0.05);与0 mg/mL组相比,0.4 mg/mL组、5 mg/mL组线虫平均寿命以及死亡率均有所下降(P<0.05);各组间A30P线虫生长发育相比无差异(P>0.05);与0 mg/mL组相比,1.0 mg/mL组、2.5 mg/mL组线虫排卵量无差异(P>0.05),与0 mg/mL组相比,0.4 mg/mL组、5 mg/mL组线虫排卵量有所下降(P<0.05);与0 mg/mL组相比,0.4 mg/mL组、5 mg/mL组线虫gst-4、gst-7、Skn-1、hsp-16.2基因水平无差异(P>0.05);与0 mg/mL组相比,1.0 mg/mL组、2.5 mg/mL组线虫gst-4、gst-7、Skn-1、hsp-16.2基因水平有所上升(P<0.05);与0 mg/mL组相比,0.4 mg/mL组、5 mg/mL组线虫弯曲次数无差异(P>0.05);与0 mg/mL组相比,1.0 mg/mL组、2.5 mg/mL组线虫弯曲次数有所上升(P<0.05);相较于与0 mg/mL组,0.4 mg/mL组、1.0 mg/mL组、2.5 mg/mL组、5 mg/mL组树突计分以及树突恢复比率均有所上升,其中树突计分以2.5 mg/mL组最优,树突恢复比率以5 mg/mL组最优(P<0.05)。结论 通过对A30P线虫应用鹿茸提取物发现,1.0 mg/mL、2.5 mg/mL两种剂量对A30P线虫生存率、生长发育、生殖能力、树突计分、抗氧化能力的能力较好,而5 mg/mL剂量下对A30P线虫树突恢复比率的效果最优。
Objective Explore the intervention effect and pharmacological effects of deer antler extract on A30P nematode.Methods Purchase A30P nematodes and divide them into 0,0.4,1.0,2.5,and 5mg/mL groups according to different concentrations of deer antler extract.Analyze the survival rate,growth and development,reproductive ability,and antioxidant capacity of A30P nematodes under different concentrations of deer antler extract,and evaluate the pharmacological effects of deer antler extract on A30P nematode movement behavior and dendritic recovery at the same concentration.Results Compared with the 0 mg/mL group,the average lifespan and mortality rate of nematodes in the 1.0 mg/mL and 2.5 mg/mL groups(P>0.05);Compared with the 0 mg/mL group,the average lifespan and mortality rate of nematodes in the 0.4 mg/mL and 5 mg/mL groups decreased(P<0.05);Comparison of growth and development of A30P nematodes among different groups(P>0.05);Compared with the 0 mg/mL group,the nematode ovulation levels in the 1.0 mg/mL and 2.5 mg/mL groups(P>0.05)were higher.However,compared with the 0 mg/mL group,the nematode ovulation levels in the 0.4 mg/mL and 5 mg/mL groups decreased(P<0.05);Compared with the 0 mg/mL group,the gene levels of gst-4,gst-7,Skn-1,and hsp-16.2 in nematodes in the 0.4 mg/mL and 5 mg/mL groups were significantly higher(P>0.05);Compared with the 0 mg/mL group,the gene levels of gst-4,gst-7,Skn-1,and hsp-16.2 in nematodes increased in the 1.0 mg/mL and 2.5 mg/mL groups(P<0.05);Compared with the 0 mg/mL group,the number of nematode bends in the 0.4 mg/mL and 5 mg/mL groups(P>0.05);Compared with the 0 mg/mL group,the number of nematode bends increased in the 1.0 mg/mL and 2.5 mg/mL groups(P<0.05);Compared to the 0 mg/mL group,the 0.4 mg/mL group,1.0 mg/mL group,2.5 mg/mL group,and 5 mg/mL group all showed an increase in dendritic score and dendritic recovery rate.Among them,the 2.5 mg/mL group had the best dendritic score,and the 5 mg/mL group had the best dendritic recovery rate(P<0.05).Conclusion Through the application of deer antler extract to A30P nematodes,it was found that two doses of 1.0 mg/mL and 2.5 mg/mL had better effects on the survival rate,growth and development,reproductive ability,dendritic scoring,and antioxidant capacity of A30P nematodes,while the best effect was observed at a dose of 5 mg/mL on the dendritic recovery rate of A30P nematodes.
