摘要
目的观察枸杞多糖(lyceum barbarum polysaccharides,LBP)干预对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病(diabetes mellitus,DM)大鼠视网膜病理改变的影响,探讨枸杞多糖对糖尿病视网膜病变的保护作用及机制。方法 SD大鼠45只,随机分为正常对照组(CON组)和糖尿病组(DM组),糖尿病组再分为模型组和LBP组。腹腔注射STZ建立DM模型,成模后LBP组予以LBP灌胃250 mg·(kg·d)-1,模型组等量生理盐水灌胃,直至处死前1d。给药后继续饲养至4、12、20周分别测血糖后处死取材取大鼠双眼眼球。用免疫组织化学染色检测各组大鼠视网膜Occludin的表达情况。结果 STZ腹腔注射大鼠成模率100%,4、12、20周测血糖浓度,CON组大鼠血糖浓度始终低于6.00 mmol·L-1;模型组血糖浓度持续增高于CON组和LBP组(P<0.01);而LBP组在同时间点血糖浓度均明显低于模型组(P<0.01)。组织学HE染色检查显示,4、12、20周时,CON组大鼠视网膜各层结构正常。随着糖尿病病程延长,模型组大鼠视网膜病理损害明显加重,而同期LBP组大鼠视网膜病理损害均轻于模型组。12、20周时模型组大鼠视网膜Occludin的表达与CON组比较升高(P<0.01)。而LBP组大鼠在第4、12周时其视网膜的Occludin表达量与CON组比较差异均无统计学意义(P>0.05),20周时Occludin表达量高于CON组(P<0.05),在12、20周时Occludin表达量均低于模型组(P<0.05)。结论 LBP可显著降低糖尿病大鼠血糖浓度,能够有效的改善视网膜各层组织病理损伤,上调Occludin表达量,稳定视网膜微血管内皮的紧密连接,抑制糖尿病血-视网膜屏障的破坏,从而发挥对糖尿病视网膜的保护作用。
Objective To observe pathological change effect of lyceum barbarum polysaccharide on retina in diabetic rats and to the protective effect of lyceum barbarum polysaccharide on retina. Methods Forty-five adult SD rats were randomly divided into normal group( CON group) and diabetes group( DM group). Diabetes group were randomly divided into model group and LBP treatment group( LBP group). Diabetic model was established through injection vena caudalis streptozotoc. LBP was fed in LBP group while an equal volume of normal sodium was administrated in model group,until the day before being killed. The three groups of rats were reared for 4 weeks,12 weeks,and 20 weeks and given blood glucose measured,then killed to take its eyes eyeballs. Retina pathological change was observed in rat retinas using HE staining and expression of Occludin was measured in rat retinas using immunohistochemistry. Results Establishment of diabetes model was100%. To measure blood sugar levels,blood glucose concentration in CON rats was always lower than 6. 00mmol·L-1at the 4th,12thand 20thweek. Difference of blood glucose concentration among model group,CON group and LBP group showed significant( P < 0. 01) while blood glucose concentrations of LBP group at the same time point were significantly lower than those of the model group( P < 0. 01). Histological HE staining in the retina of diabetic rats showed that retinal had no significant anomalies in CON rats at all time points.With the extended duration of diabetes,retinal pathological damage significantly increased in model rats while retinal pathologic lesions of the LBP group were lighter than those of the model group. Retinal retinal immunohistochemistry staining showed: retinal Occludin expression significantly decreased with the duration of the extension in CON group( P < 0. 01). The Occluding expression increased significantly in LBP group at 20week compared with the CON group( P < 0. 05) while it decreased significantly in LBP group at 4week and 12week compared with the model group( P < 0. 05). Conclusion LBP can reduce the blood glucose concentration in diabetic rats and improve the pathological injury of retina effectively. LBP has a protective effect on the destruction of blood-retinal barrier in diabetic rats.
