摘要
目的系统评价鼠双微染色体2基因(MDM2)基因和抑癌基因p53在脑胶质瘤的表达及其对肿瘤发生发展的影响。方法从国内外数据库检索关于MDM2和p53的表达与胶质瘤的关系的对照研究,符合条件的研究结果采用Revman5.3软件以优势比(OR)和95%置信区间(CI)为效应指标进行Meta分析。结果最终纳入文献10篇,Meta分析的结果显示:胶质瘤和对照组中p53表达的差异(OR=29.0;95%CI=10.63—79.13;P〈0.01)I、胶质瘤和对照组中MDM2表达的差异(OR=24.83;95%CI=8.86—69.63;P〈0.01)、p53表达情况与胶质瘤病理分级关系的表达差异(OR=0.26;95%CI=0.18—0.38;P〈0.01)、MDM2表达情况与胶质瘤病理分级关系的表达差异(OR=0.52;95%CI=0.35—0.77;P〈0.01)均有统计学意义。结论p53与MDM2表达异常与脑胶质瘤发生、发展过程中密切相关。对脑胶质瘤中MDM2、p53表达的检测分析将有利于进一步探讨脑胶质瘤发生发展的分子机制,以期为临床的预防、诊断和治疗提供理论依据。
Objective The expression of Murine double mimute 2 (MDM2) and p53 in glioma and its association with the occurrence and development were discussed. Methods Odds ratio (OR) and 95% confidence intervals ( 95% CI) were determined for expression of MDM2 and p53 using frxed or random model with Revman 5.3. Results Evemually 10 case-control studies were enrolled. Meta-analysis effect showed that MDM2 and p53 expression differences of each group were all statistically significant. Conclusion Abnormal expression of MDM2 protein and p53 gene in glioma is closely related to the occurrence and development of glioma. Detection and analysis of the expression of MDM2 and p53 in glioma will be helpful for the research of molecular mechanism of glioma development in order to provide theoretical basis for prevention, clinical diagnosis and treatment.
