摘要
为考察珠蓼散的提取工艺参数、抑菌效果以及对仔猪腹泻的防治作用,以便为其临床应用提供试验依据,采用正交试验设计优化珠蓼散的最佳提取工艺参数,采用纸片法和牛津杯法测定珠蓼散对大肠杆菌和沙门氏菌的抑菌效果,最后选用50头健康和40头同期发生腹泻三元杂交仔猪分别进行了预防和治疗试验。结果表明,珠蓼散的收膏率最高可达17%;纸片法测得珠蓼散大肠杆菌的抑菌圈直径为11.0-14.5 mm,对沙门氏菌的抑菌圈直径为10.0-13.0 mm;牛津杯法测得珠蓼散对大肠杆菌的抑菌圈直径为16.0-17.0 mm,对沙门氏菌的抑菌圈直径为14.0-15.0 mm;预防试验中,试验组较空白对照组发病率降低80%,较西药对照组降低20%,且临床症状轻;治疗试验中,所有试验组的治愈率均为100%,较硫酸庆大霉素提高10%。珠蓼散的最佳提取工艺参数为:提取时间1.5 h,提取3次和10倍水;对大肠杆菌和沙门氏菌均有较好的体外抑制效果;对细菌性仔猪腹泻具良好的预防和治疗作用。
In order to study the extraction process, antibacterial effect, prevention and treatment effect on diarrhea of Zhuliao powder, so as to provide experimental basis for its clinical application, orthogonal design was adopted to optimize its process parameters; antibacterial paper method and oxford cup method were used to determine the diameter of the inhibition zone; and finally,prevention test on 50 healthy crossbred piglets and treatment test on 40 diarrhea piglets at the same time were conducted. Results showed that the yield rate of extract in group Ⅸ was the highest of 17%; with the antibacterial paper method, the diameter of inhibition zone of E. coli was 11.0-14.5 mm, that of Salmonella was 10.0-13.0 mm; and with the oxford cup antibacterial method, the diameter of nhibition zone of E. coli was 16.0-17.0 mm, that of Salmonella was 14.0-15.0 mm; in prevention test, the diarrhea incidence in trial group was decreased by 80% than the control group and 20% than the western medicine group; in treatment test, the cure rates of all trial groups were 100%, which was 10% higher than that of Gentamicin sulfate group. It is prompted that the best extract process parameters of Zhuliao powder were： 1 hour of extract time, 3 times extract and 12 times of water; Zhuliao powder had good antibacterial effect on E. coli and Salmonella, as well as it can effectively prevent and cure piglets＇ bacterial diarrhea.
