桑叶黄酮对鸡白痢沙门氏菌的体外抑菌效果试验

摘要 通过测定最小抑菌浓度、最小杀菌浓度和抑菌圈试验等方法,评价桑叶黄酮对鸡白痢沙门氏菌的抑制效果。用扫描电镜观察、黏附试验和RT-qPCR测定T3SS毒力基因表达水平等方法,探讨桑叶黄酮对鸡白痢沙门氏菌的体外抑制效果及其机理。结果表明,桑叶黄酮对鸡白痢沙门氏菌的最小抑菌浓度和最小杀菌浓度均为25 mg/mL;扫描电镜可见高浓度桑叶黄酮处理后的鸡白痢沙门氏菌菌体多裂解,孔洞明显增多。黏附试验显示,较高浓度桑叶黄酮能显著降低鸡白痢沙门氏菌对鸡胚成纤维细胞的黏附能力。RT-qPCR结果显示,桑叶黄酮可极显著提高T3SS毒力基因prgK、invA的表达水平(P<0.01),显著提高prgI的表达水平(P<0.05)。说明桑叶黄酮在体外能有效抑制鸡白痢沙门氏菌的增殖,破坏菌体表面形态,降低黏附能力;低浓度桑叶黄酮会刺激T3SS毒力基因表达,提高其侵袭能力。研究结果为桑叶天然活性物质抑菌功效的开发及其在家禽养殖行业的应用提供了科学证据。 This study evaluated the inhibition effect of mulberry leaf flavonoids on Salmonella pullorum by measuring the minimum inhibitory concentration,minimum bactericidal concentration and conducting inhibition zone experiments.This study used scanning electron microscopy observation,adhesion experiments and RT-qPCR determining the expression level of T3SS genes,aiming to discuss the inhibitory effect and its mechanism in vitro of mulberry leaf flavonoids on Salmonella pullorum.The results showed that both minimum inhibitory concentration and minimum bactericidal concentration of mulberry leaf flavonoids on Salmonella pullorum were 25 mg/mL.Scanning electron microscopy showed that most of the Salmonella pullorum treated with high concentration mulberry leaf flavonoids were lysed,and the number of pores was significantly increased.Adhesion experiment showed that high concentration of mulberry leaf flavonoids could significantly reduce the adhesion ability of Salmonella pullorum to chicken embryo fibroblasts.RT-qPCR showed that mulberry leaf flavonoids could high-significantly increase expression levels of prgK and invA(P<0.01),and significantly increase expression level of prgI(P<0.05)in T3SS genes.In conclusion,mulberry leaf flavonoids can effectively inhibit the proliferation of Salmonella pullorum in vitro,disrupt the surface morphology of the bacterial cells,and reduce adhesion ability.Low concentration mulberry leaf flavonoids can stimulate expression of T3SS virulence genes and enhance their invasive ability.This study s results provide scientific evidence for the development of inhibitory effects of natural active substances in mulberry leaf and their application in livestock and poultry industry.
