槐果碱的体外抗弓形虫作用 被引量:2

摘要 [目的]评估槐果碱的体外抗弓形虫作用.[方法]采用CCK-8法测定槐果碱对HeLa细胞的毒性和对弓形虫选择性作用的效果,光学显微镜下观察细胞形态变化,应用台盼蓝拒染法检测活细胞百分比.[结果]槐果碱浓度为500μmol/L时,对HeLa细胞有明显的抑制作用;槐果碱体外抗弓形虫作用的选择性指数为0.96,效果低于乙胺嘧啶(1.77),但优于螺旋霉素(0.73);给予200μmol/L槐果碱治疗24 h后,HeLa细胞破损程度明显降低,细胞内外速殖子数量显著减少,且在10~200μmol/L浓度范围内时活细胞比例随给药浓度的增加而明显上升.[结论]槐果碱在体外表现出良好的抗弓形虫活性,且对弓形虫感染的HeLa细胞具有一定程度的保护作用. OBJECTIVETo evaluate the effects of sophocarpine for anti-toxoplasma gondii tachyzoites in vitro.METHODS The cytotoxicity and the selective anti-toxoplasma effect of sophocarpine were detected by CCK-8assay,the morphological change of HeLa cells were observed by light microscopy,and the percentage of living cells was detected by using trypan blue exclusion test.RESULTS The sophocarpine with ≤200μmol/L concentrations had obvious inhibiting effect for HeLa cells,and the selectivity index(SI)of sophocarpine on anti-toxoplasma was 0.96,which was lower than pyrimethamine but superior than spiramycin(SI=1.77 and 0.73,respectively).After exposure to 200μmol/L sophocarpine for 24 h,the morphology of HeLa cells restored remarkably and the number of tachyzoites decreased obviously,furthermore,the sophocarpine in 10-20 μmol/L concentrations improved the percentage of living cells in a concentration-dependent manner.CONCLUSION The sophocarpine has good anti-toxoplasma gondii activity with a certain degree of protection to toxoplasma-infected HeLa cells.
