摘要
通过富集培养、稀释涂布、平板划线等方法,从菌菇栽培基质中分离筛选到一株纤维素酶产量较高的菌株FQZ6,根据菌落形态及镜检初步判定该菌株为放线菌属(Actinomycetes);并对其产酶条件进行研究.结果表明,菌株产纤维素酶的最佳发酵条件为:基质为2 g·L-1麸皮,氮源为3 g·L-1明胶,培养时间72 h,10%接种量,在此条件下,菌株FQZ6的酶活达到190.14 U·m L-1.该菌株纤维素酶的最适温度和p H值分别为40℃和8,Mg2+、K+、Fe3+离子对纤维素酶活性有一定的激活作用,Mn2+、Zn2+、Ca2+对纤维素酶影响不大,而Cu2+、Hg2+对纤维素酶活性有抑制作用.
The strain FQZ6 isolated from fungus mushroom cultivation matrix was identified as Actino- mycetes according to the colony morphology and microscopy by enrichment culture, dilution method such as coating, plate marking. And the enzyme production conditions were studied. The results showed that the best fermentation conditions for strain were matrix of 2 g·L^-1 bran, nitrogen source for 3 g·L^-1 gelatin, 72 h of incubation time, quantity of 10%, in this condition, the enzyme activity of strain FQZ6 190.14 U ·mL^-1. The optimum temperature and pH value of cellulase were 40℃ and 8, respectively. Mg^2+、K^+、Fe^3+ could activate the cellulase, but Mn^2+、Zn^2+、Ca^2+ had little im- pact. While Cu^2+、Hg^2+ had only modest inhibitory effect.
