摘要
以同源四倍体台湾泡桐叶片和叶柄为外植体,将其分别置于含有不同植物生长调节剂及其不同质量浓度(0.1~1.1 mg/L NAA和6~20 mg/L 6-BA)的组合培养基上诱导再生植株,筛选最佳外植体和最适培养基,建立体外植株再生系统。结果表明,同源四倍体台湾泡桐体外植株再生的最佳外植体是叶片;其愈伤组织诱导的最适培养基为MS+0.1 mg/L NAA+14 mg/L 6-BA;芽分化的最适培养基为MS+0.3 mg/L NAA+16 mg/L 6-BA;幼芽生根的最适培养基为1/2MS+0.1 mg/L NAA。
The leaves and petiole of autotetraploid Paulownia kawakamii were placed on media with different hormones and concentrations(0. 1--1. 1 mg/L NAA,6--20 mg/L 6-BA) , so as to screen the optimal explants and media,and establish the plantlet regeneration system in vitro. The results indicated that the leaves were the optimal explants for plantlet regeneration of autotetraploid Paulownia kawakamii in vitro. Moreover,the optimal media for callus induction from Paulownia kawakamii was MS + 0.1 mg/L NAA + 14 mg/L 6-BA,the optimal media for shoot differentiation was MS + 0.3 mg/L NAA + 16 mg/L 6-BA, and the optimal rooting media was 1/2MS +0.1 mg/L NAA.
