多粘类芽孢杆菌HD-1产纤维素酶的条件优化 被引量:11

Optimization of producing conditions of the cellulase from Paenibacillus polymyxa HD-1

摘要 采用DNS比色法,通过单因素实验和正交实验对多粘类芽孢杆菌HD-1产纤维素酶的培养基主要成分和培养条件进行优化。结果表明:该菌种产纤维素酶的最适培养基为麸皮0.5%、酵母膏1.25%、磷酸二氢钾0.04%、硫酸镁0.1%,培养基初始p H8.0;最适培养条件为接种量4%,培养温度35℃,培养时间51h。在此优化条件下,多粘类芽孢杆菌产纤维素酶活性可达88.3U/m L。 In order to improve the production of cellulase,the fermentation medium and culture conditions for Paenibacillus polymyxa HD-1 were optimized by single factor test and orthogonal test. The results showed that the best compositions of fermentation medium were 0.5% of bran, 1.25% of yeast extract,0,04% of KH2PO4.3H20, 0.1% of MgSO4.7H20,with the initial pH of the medium was 8.0;and the best fermentation inoculum size was 4% ,culture temperature at 35℃ and culture time for 51 hours. Under this optimal conditions,the activity of cellulase from Paenibacillus polymyxa HD-1 could reach 88.3U/mL.
