摘要
目的探讨细胞周期调控因子p21在皮肤病理性瘢痕和瘢痕癌组织中的作用。方法 1采用激光扫描共聚焦显微技术,对瘢痕癌和病理性瘢痕石蜡切片组织进行p21免疫荧光双标记,同时采集三维立体图像;2应用免疫组织化学染色(SP)法,检测瘢痕癌组织、病理性瘢痕被覆上皮和正常皮肤表皮3组组织中p21蛋白的表达;3采用核酸分子原位杂交技术检测p21m RNA在3组组织中的表达;4对各项指标进行相关性分析,计算平均光密度和阳性面积,并结合图像分析,所有数据运用SPSS 17.0软件包进行统计学分析。结果 1免疫荧光双标记结果:p21蛋白在瘢痕癌癌组织中呈较强荧光,为高表达;在病理性瘢痕上皮中呈微弱荧光,为阴性表达;2免疫组织化学和原位杂交结果:p21蛋白和p21m RNA在瘢痕癌癌组织中呈高表达,在病理性瘢痕上皮中呈低表达,在正常皮肤表皮呈阴性表达。瘢痕癌组表达强度(平均光密度)和表达水平(阳性面积)分别与病理性瘢痕及正常皮肤组相比较,差异均有统计学意义(P<0.01);但病理性瘢痕组与正常皮肤组比较,差异无统计学意义(P>0.05);3相关性分析:在瘢痕癌组织中,p21蛋白与p21m RNA的表达呈正相关。结论瘢痕癌中细胞周期调控因子p21的过表达,其凋亡抑制调控作用有待进一步探讨,p21的表达可能具有组织特异性。
【Objective】 To investigate the expression and signification of cell cycle control genes p21 in scar cancer.【Method】① The paraffin sections of the skin pathological scar and scar cancer tissues were double labeled with p21 by using immunofluorescence and scanned by the laser scanning confocal microscopy to form the three-dimensional images. ②The expression of p21 protein was detected in the scar cancer tissue, the pathological scar epithelium and normal skin epidermis respectively by immunohistochemical staining(SP). ③The expression of p21 mRNA was detected by nucleic acid molecules in situ hybridization. ④ The average optical density and positive area were analyzed by SPSS 17.0 software package for statistical analysis. 【Results】① Double-labeling immunoflurescence results: p21 protein showed strong fluorescence in the scar cancer tissues and weak fluorescence in the pathological scars epithelium.② Immunohistochemical and in situ hybridization results: p21 protein and p21 mRNA showed high expression in the scar cancer, but low expression in the pathological scar epithelium and negative expression in the normal skin epidermis. The mean optical density and the positive area in the scar cancer had significant difference compared with pathological scar group and normal skin group(P〈0.01), but there were not statistically difference in the mean optical density and the positive area between the pathological scar group and normal skin group(P〈0.05). ③ Correlation analysis: The expression of p21 protein and p21 mRNA showed positive correlation in the scar cancer tissue. 【Conclusion】The overexpression of cell cycle control genes p21 in the scar cancer and its role of inhibition of apoptosis is to be further explored. The expression of p21 may have tissue-specific.
