摘要
目的 采用HPLC法测定不同产地杜仲雄花中5种活性成分的量.方法 采用Thermal hypersil (250 mm×4.6 mm,5μm)色谱柱,以甲醇(B)-0.5%磷酸水溶液(C)为流动相,梯度洗脱;检测波长为206 nm(0 ~15 min),236 nm (15 ~55 min);体积流量1.0 mL/min.结果 桃叶珊瑚苷、京尼平苷酸、绿原酸、车叶草苷、京尼平苷分别在0.395 ~ 11.85 μg、0.481 ~ 14.43 μg、0.416 ~ 12.48 μg、0.48~14.4 μg、0.481 ~ 14.43 μg范围内线性关系良好,平均加样回收率为97.5% ~ 98.3%.结论 该方法简便快捷重复性好,可用于杜仲雄花的质量控制.
AIM To simultaneously determine five bioactive compounds (aucubin, geniposidic acid, chloro- genic acid, asperuloside and geniposide) by HPLC. METHODS Samples were separated on a Thermal hypersil gold column (250 mm ×4.6 mm,5μm) and detected by an ultraviolet detector. The UV wavelength was set at 206 (0- 15 min), 236 nm( 15 - 55 min). Mobile phase consisted of methanol (B) and phosphoric acid-water (0. 5% ) (C) using a gradient elution. Analytes were performed at 30 ℃ with a flow rate of 1.0 mL/min. RE- SULTS In quantitative analysis, the five components showed good regression (r2 〉 0. 999 9) within linear ran- ges, and their average recoveries rate were in the range of 97.5% - 98.3%. CONCLUSION This study will provide methodological reference for the quality control of male flowers of E. Ulmoides.
