摘要
用K-卡拉胶包埋啤酒酵母制成固定化细胞,在0.05mol/L葡萄糖,0.022mol/L腺苷酸,0.012mol/LMgSO_4·7H_2O,0.2mol/L氯化钾,0.2mol/L磷酸钾缓冲溶液(pH7.0)中,35℃反应4小时,5′-三磷酸腺苷转化率可达42%,在50ml三角烧瓶中分批反应,连续使用5次,5′-三磷酸腺苷转化率仍维持在37%以上。
The immobilized beer yeast cells entrapped in K-carrageenan gel beads were used to biosynthesize adenosine 5′-triphosphate from a buffer (pH 7.0) substrate solution containing 0.05 mol/L glucose, 0.022 mol/L AMP, 0.012 mol/L MgSO_4·7_2O, 0.2 mol/L KCl, and 0.2 mol/L potassium phosphate. The biosynthesis reaction temperature is 35℃, the reaction time being 4 hours; the conversion rate of ATP can reach 42%. When the reaction wasrepeated batch wise in a 50ml conical flask, the conversion rate of ATP can remain above 37% after five succesive times.
