摘要
用开孔明胶包埋啤酒酵母制成固定化细胞,在含15g/L单磷酸腺苷、150mmol/L葡萄糖、16mmol/L硫酸镁的250mmol/L磷酸二氢钠缓冲液(pH7.0)中35℃振荡反应2h,生产三磷酸腺苷(ATP)的转化率可达95%。在前一批底物溶液灭菌和添加烟酰胺腺嘌呤二核苷酸的条件下,固定化酵母细胞可反复使用10次,ATP转化率稳定维持在90%以上。
The immobilized Saccharomyces cerevisiae cells entrapped in open pore gelatin matrix were used to biosynthesize ATP with a substrate solution of 15g/L AMP and 150mmol/L glucose in 250mmol/L NaH2PO4 buffer solution (containing 16mmol/L MgSO4, pH 7.0). The conversion rate of ATP could roach to 95% at 35℃ within 2h. After sterilization and addition of NAD^+ in the used substrate solution, the immobilized cells could be recycled for 10 times to maintain the conversion rate of ATP over 90%.
