摘要
目的 :观察生肌化瘀方及拆方对创面成纤维细胞的α1(Ⅰ )mRNA和α1(Ⅲ )mRNA表达的影响 ,探讨其促进创面修复呈皮肤修复的作用机理。方法 :采用体外培养创面肉芽组织成纤维细胞 ,以乳鼠皮肤成纤维细胞作对照 ,分别加入生肌方、化瘀方及生肌化瘀方大、小剂量药物血清 ,运用NthernBlot分子杂交法检测成纤维细胞Ⅰ、Ⅲ前胶原mRNA的表达。结果 :生肌方能够加强创面成纤维细胞Ⅰ、Ⅲ型前胶原mRNA的表达 ,高于模型组 ;化瘀方能够降低创面成纤维细胞Ⅰ、Ⅲ型前胶原mRNA的表达 ,低于模型组 ;生肌化瘀方各剂量组Ⅰ、Ⅲ型前胶原mRNA的表达与正常组相比差异均无显著性。结论 :生肌化瘀方通过调控创面成纤维细胞的α1(Ⅰ )mRNA和α1(Ⅲ )
Objective:The study was aimed at investigating the mechanism of the three Traditional Chinese Medicine (TCM) formula,(Shengji Fang,Huayu Fang and Shengji Huayu Fang) in accelerating wound healing into skin restoration at the cell and gene level.Method:The fibroblasts from granulation tissue in wound were cultured in vitro and compared with the normal skin fibroblast of suckling rat, which were incorporated with serum containing Shengji Fang,Huayu Fang,small and high doses of drug Shengji Huayu Fang prepared by serum pharmacology repectively.the mRNA expression of types ⅠandⅢcollagens of fibroblasts were assessed by Northern blot hybridization.Result:Shengji Fang could promote the proliferation of fibroblasts and enhance the expression of α 1(Ⅰ) and α 1 (Ⅲ) In contrast,Huayu Fang could inhibit the expression,with significantly different from the control model fibroblasts.Finally,Shengji Fang Huayu Fang has bidirectional regulation upon the two parameters.Conclusion:The TCM formula could accelerate wound healing into skin restoration through promoting the proliferation of fibroblasts from granulation tissue of wound,regulate the expression of Ⅰ、Ⅲtype procollagen mRNA.
