摘要
目的:评价曲安奈德对瘢痕疙瘩成纤维细胞增殖及Ⅰ、Ⅲ型前胶原mRNA表达的影响。方法:取人瘢痕疙瘩及正常皮肤组织培养的第58代的成纤维细胞,在含有不同浓度的曲安奈德的环境中培养。应用噻唑蓝(MTT)法检测细胞增殖、RT-PCR检测Ⅰ、Ⅲ型前胶原mRNA表达。结果:浓度为0.05~1.0g/L的曲安奈德能明显抑制瘢痕疙瘩和正常皮肤成纤维细胞的增殖;浓度为0.10~1.0g/L的曲安奈德能降低瘢痕疙瘩和正常皮肤成纤维细胞Ⅰ、Ⅲ型前胶原mRNA表达。所有的抑制作用在浓度1g/L时达到最高。结论:曲安奈德对瘢痕疙瘩和正常皮肤成纤维细胞的增殖及Ⅰ、Ⅲ型前胶原mRNA表达有明显的抑制作用。
Objective: To assess the impact of triameinolone acetonide on the proliferation and expression of Ⅰ, Ⅲ procollagen mRNA in human keloid fibroblasts. Methods: Triamcinolone acetonide of different concentration was added to cultured fibroblasts for 5 - 8th generations of passage from human keloid and normal skin tissue. Proliferation activity of fibroblasts was examined using MTF and RT - PCR assay to analyze the expression of I and III procollagen mRNA. Results: Proliferation of fibroblast from the keloids and normal skin were inhibited significantly. The concentration of 0.05 - 1.0 g/L triamcinolone acetonide was markedly inhibited the proliferation of fibroblasts and the concentration of 0.10 ~ 1.0 g/L triamcinolone acetonide decreased the expression of I, III procollagen mRNA in fibroblasts cultured from keloid and normal skin. The inhibitive effects were the most high when 1 g/L triamcinolone acetonide added. Conclusion: Triamcinolone acetonide can markedly inhibit the proliferation and the expression of I and III procollagen mRNA of fibroblasts from keloids and normal skin.
