Expression of Fas, FasL, Bcl-2 and Bax in acute cardiac allograft rejection in rats

摘要 目的 :探讨Fas、FasL、Bcl 2和Bax在大鼠心脏移植急性排斥反应中的表达及意义。方法 :进行大鼠心脏移植 ,原位末端标记 (TUNEL)技术检测移植心的凋亡细胞 ,免疫组织化学方法检测Fas、FasL、Bcl 2和Bax的表达并用计算机图像分析定量测定。结果 :随着术后天数增多 ,排斥反应分级上升 ,细胞凋亡增加 ,FasL表达升高 ,Bcl 2表达下降而Fas、Bax无明显变化 ,AI于第 7d达高峰 (11 4 6± 5 35 ) ,FasL上升第 5d达高峰 (32 17± 8 15 ) ,Bcl 2第 5d下降最低 (8 79± 1 98)。结论 :FasL和Bcl 2与心脏移植急性排斥及组织损伤密切相关 ,FasL、Bcl 2的检测可作为急性排斥的监测指标。 Aim: To investigate the expression of Fas, FasL, Bcl 2 and Bax and their significance in acute cardiac allograft rejection in rats. Methods:Groups of Wistar rats underwent heterotopic heart transplantation from allogeneic SD rats. The cardiac grafts were harvested at 1, 3, 5, or 7 days after transplantation. The normal and transplanted hearts were detected for apoptotic cell death and expression of Fas, FasL, Bcl 2 and Bax using TUNEL and immunohistochemical method. Histopathological rejection grade and AI were analyzed. Quantitative immunohistochemical measurement was performed with computer imaging analysis. Results:With the posttransplantation day increased, the rejection grade and AI elevated, the expression of FasL increased, Bcl 2 decreased, Fas and Bax had few change. The peak of AI, FasL expressing and the bottom of Bcl 2 expressing were 7th, 5th and 5th posttransplantation day respectively and the value was 11.46±5.35, 32.17±8.15 and 8.79±1.98 severally. Conclusion:FasL and Bcl 2 are highly related with acute cardiac allograft rejection and tissue injury in rats, FasL and Bcl 2 may be used as a monitoring index in acute rejection.
