bcl-2 mRNA在正常和增生前列腺组织中的表达及意义 被引量:2

EXPRESSION OF bcl 2 mRNA IN NORMAL AND HYPERPLASTIC PROSTATE TISSUES

摘要 目的:探讨bcl2mRNA在良性前列腺增生发病过程中所起的作用。方法:采用斑点杂交技术研究正常和增生前列腺组织中bcl2mRNA表达情况,并加以比较,同时采用原位杂交技术了解bcl2mRNA在增生前列腺中表达。结果:bcl2mRNA在增生前列腺组织中表达明显高于在正常前列腺组织中的表达,增生组织中平均表达值是正常组织的(1.71±0.72)倍;原位杂交显示:bcl2mRNA主要在前列腺上皮组织中表达,并呈现由基底细胞向表层上皮细胞表达逐渐减弱趋势。结论:bcl2mRNA在增生组织中高表达表明增生前列腺上皮细胞抗凋亡作用强于正常组织,提示bcl2参与前列腺细胞增殖与凋亡过程,增生前列腺中bcl2高表达可能是诱发良性前列腺增生的重要病因。 Objective: To explore the effect of bcl 2 mRNA on pathogenesis of benign prostatic hyperplasia (BPH). Methods: Gene expression of bcl 2 was studied in normal and hyperplastic prostate tissues by means of dot blot hybridization technique, and in BPH tissues by using in situ hybridization technique. Results: The mRNA levels of bcl 2 in BPH tissues were significantly higher than those in normal prostate, and the average dot IOD value ratio of BPH to normal prostate was ( 1.71 ±0.72) ( P <0,01). bcl 2 mRNA was located mainly in the epithelium of BPH tissues, and the positive expression rate of epithelia cells and their degree were higher in basal cells than that in superficial cells. Conclusion: Antiapoptosis power in epithelia cells of BPH was higher than that in the normal. The results suggest that bcl 2 mRNA affects the balance between prostatic cell proliferation and apoptosis, and bcl 2 might play an important role in the development of BPH.
