摘要
目的初步探讨烟酰胺对紫外线 (UV)所致光老化皮肤真皮羟脯氨酸(HP)含量是否有影响。方法利用无毛昆明种小鼠动物模型模拟慢性长期紫外线照射所致的皮肤光老化 ,采用UVB和UVA混合光源 ,经17周剂量由1MED渐增至3MED的紫外线照射背部皮肤 ,然后观察15周 ,测定真皮HP含量。分组方法 :非紫外线照射组 (不施加任何处理因素 ) ;烟酰胺组 (照射紫外线结束30min后 ,0.2mol/L烟酰胺丙酮溶液200μl涂抹于小鼠背部皮肤 ,每周2次 ) ;丙酮对照组 (照射紫外线30min后 ,90%丙酮溶液200μl涂抹小鼠背部皮肤 ,每周2次 ) ;紫外线照射组 (仅照射紫外线 )。结果小鼠背部皮肤HP含量烟酰胺组明显高于紫外线照射组和丙酮对照组(P<0 01)。各组腹部真皮HP含量差异均无显著性(P>0 05)。非紫外线照射组、烟酰胺组背、腹真皮HP含量自身比较差异均无显著性 ,而紫外线照射组、丙酮对照组背、腹真皮HP含量差异有显著性 (P<0.01) ,腹部明显高于背部。结论皮肤涂抹烟酰胺可抵抗由皮肤光老化引起真皮HP含量降低。
Objective To preliminarily explore whether nicotinamide could effect the levels of hydroxyproline (HP) in dermis of photoaging skin induced by ultraviolet radiation Methods The photoaging of back skin induced by chronic and long term exposure to ultraviolet radiation with animal model of hairless Kunming mice The hairless mice were continuously exposed to a lamp with a mixed light spectrum of ultraviolet B and ultraviolet A at increasing doses from 1 MED to 3 MED for 17 weeks The levels of HP in dermis of mice were determined at the 15th week after the end of the 17 week exposure to ultraviolet radiation The mice were randomly divided into 4 groups, such as non_ultraviolet radiation group without any treatment, nicotinamide group, the back skin of which painted by 200 μl 0.2 mol/L nicotinamide acetone solution after 30_miunte ultraviolet radiation twice a week, acetone control group, the back skin of which was painted by 200 μl 90% acetone solution after 30_minute ultraviolet radiation twice a week, ultraviolet group only radiated by ultraviolet ray. Results The significantly higher levels of dermis HP were observed in mice backs of nicotinamide+ultraviolet radiation group compared with those in ultraviolet radiation group and acetone control group (P<0 01) There were no significant differences in the levels of dermis HP in abdomen of mice between these 4 groups (P>0 05) Significant difference of the levels of dermis HP between back and abdomen of mice were observed in ultraviolet radiation group and acetone control group, the levels of HP in dermis of mice abdomen of these two groups were higher compared with the mice back (P<0.01), but no significant difference were noticed in the non_ultraviolet radiation group and nicotiamide+ultraviolet radiation group. Conclusion Nicotinamide applied on the surface of skin could resist the decreasing levels of HP in dermis of skin induced by skin photoaging.
