摘要
目的 :改进大黄提取工艺 ,降低成本 ,减小毒性。方法 :大黄粉碎后 ,先用 95 %乙醇提取 ,浓缩所得浸膏再加 2 0 %硫酸和氯仿强烈搅拌下回流提取 ,分出氯仿层 ,浓缩至适当体积 ,依次用 2 5 %碳酸氢钠、 5 %碳酸氢钠和 0 5 %氢氧化钾萃取。萃取液酸化后分别得大黄酸、大黄素和芦荟大黄素粗品。结果 :在保持收率不变的条件下 ,采用该方法使氯仿和硫酸的用量均减少为原来的 1/ 8。结论 :采用该方法减少了氯仿和硫酸的用量 ,降低了成本 ,同时减少了氯仿对操作者健康的危害 。
Objective:To improve the traditional technology of extracting free anthraquinones from Rheum for reducing the production cost and decreasing the harm on workers by chloroform.Methods:In the new technology,the Rheum was extracted at first with 95% alcohol,then the alcohol was removed and the extract was refluxed for 2.5 h in 20% sulfuric acid and chloroform.The chloroform was separated from sulfuric acid and was extracted successively with 2.5% sodium bicarbonate,5% sodium carbonate and 0.5% potassium hydroxide.The solutions were acidified with 20% sulfuric acid to get respectively Rhein,Emodin and Aloeemodin.Results:When yields were remained to be unchanged,both the needs for sulfuric acid and chloroform in the new technology were decreased to 1/8 of that in the traditional technology.Conclusion:That the new technology was used decreased in need for sulfuric acid and chloroform and thus production cost was reduced and the harm resulted from chloroform to health of worker was also alleviated.The new technology can be used for extracting free anthraquinones from Rheum in laboratory.
