摘要
目的观察兔关节软骨细胞与胶原海绵在体外复合培养以及体内植入后异位成软骨情况。方法采用酶消化法分离、培养幼兔关节软骨细胞,取第三代细胞以3×107/ml密度均匀接种于胶原海绵中,分别通过倒置显微镜和扫描电镜观察软骨细胞在载体材料上生长增殖情况;将体外培养10d的软骨细胞/胶原海绵复合物植入裸鼠背部皮下,术后12周取材,进行大体观察和HE染色。结果软骨细胞均匀分布于胶原海绵的孔壁和网眼内,并可分泌基质样物;复合培养物体内植入12周后可以形成新生透明软骨组织。结论胶原海绵可以作为软骨细胞生长的支架载体,异位构建出组织工程化软骨组织。
Objective To observe the growth of rabbit chondrocytes incubated on the collagen sponge in vitro and to investigate the ectopic chondrogenesis by implantation of chondrocyte scaffold composite. Method The chondrocytes of young rabbit were isolated and cultured by enzyme digestion, the third generation of them were seeded on collagen sponge in a density of 3×107/ml. The inverted microscopy and scanning electron microscopy were employed to observed the growth and proliferation of chondrocytes on collagen sponge.After 10 days of mixed cultured, the chondrocytes combinded with collagen sponge were implanted into the dorsal subcutaneous tissue of nude mouses.The macroscopic and histological examinations of samples were performed at 12 weeks after operation. Result Chondrocytes were seen to distribute evenly inside the collagen sponge with matrix secreting in vitro and hyaline cartilage formation were observed at the 12th week postoperatively. Conclusion Collagen sponge can act as carrier of chondrocytes to construct tissue engineered cartilage in ectopia.
